Objective: To observe the effect of Qilian Wendan Decoction on the level of Cox-2 and CagA protein from mice gastric mucosa of Helicobacter pylori associated gastritis(spleen deficiency with dampness-heat syndrome),and to explore the mechanism of Qilian Wendan Decoction in treating Helicobacter pylori associated gastritis(spleen deficiency with dampness-heat syndrome),which could provides a new method for the treatment of Helicobacter pylori associated gastritis.Methods: Sixty aged 6-8 weeks Balb/c mice of SPF grade were weighed(20±1.47)g,male and female.According to the male and female each half were randomly divided into 6 groups,namely the model group,Qilian Wendan Decoction high dose group,middle dose group,low dose group,western medicine(Livzon triple)group and blank control group.In addition to the blank control group,the remaining 5 groups were all modeled by using compound factors,those were HP gastro-infection + simulated dampness-heat environment + high-fat/high-sugar diet(80% basic diet powder+12% lard+8% honey)to establish a model of H.pylori-associated gastritis with spleen-deficiency and dampness-heat syndrome.Each group was given intragastrically with the corresponding drug.The model group and the blank control group were intragastrically administrated with equal volume of normal saline for one day and once time for 14 days.After the end of the experiment,the mice were sacrificed.The eradication of Helicobacter pylori was observed by mixed staining,the pathological conditions of gastric mucosa in mice were observed by HE staining,and the expression of Cox-2 and CagA protein in mouse gastric mucosa was observed by immunohistochemistry.Results:1.The mental status,coat color,activity,food intake,water intake,defecation and other general conditions of mice in Qiliang wendan decoction groups were significantly improved compared with the model group.2.Observe the gastric mucosal pathological damage of the experimental mice and the H.pylori colonization after statistical analysis with average optical density and average area values.Among them,compared with the blank group,the mice in the model group had gastric mucosal pathological scores.The statistical analysis of H.pylori colonization with the average optical density values was higher than that of the blank group(P<0.01).The difference was statistically significant.Compared with the model group,the Qilian Wendan Decoction each dose group,the gastric mucosal pathological scores in the Livzon triple group and the statistical analysis of H.pylori colonization by mean optical density values were lower than those in the model group(P<0.05).The difference was statistically significant.3.Compared with the blank group,the expression of Cox-2 protein in the gastric mucosa of the model group was significantly increased(P<0.01).Compared with the model group,the gastric mucosa of Cox-2 in the western medicine group mice The protein content was significantly decreased,and the difference was statistically significant(P<0.01).The content of Cox-2 protein in the gastric mucosa of the high-dose group and the medium-dose group of the traditional Chinese medicine decreased,the difference was statistically significant(P<0.05).4.Compared with the blank group,the gastric mucosal CagA protein content in the model group was significantly increased(P<0.01).Compared with the model group,the Western medicine group,the traditional Chinese medicine high-dose group,the Chinese medicine medium dose The content of CagA protein in the gastric mucosa of mice in the low-dose group was significantly lower than the control group,and the difference was statistically significant(P<0.01).Conclusion:1.Qilian Wendan Decoction has a good therapeutic effect on H.pylori-associated gastritis(spleen deficiency with dampness-heat syndrome).2.Its mechanism of action may be through down regulating the expression of Cox-2 and CagA protein from gastric mucosa,inhibiting the release of inflammatory cells and reducing inflammatory damage,so as to achieve the therapeutic effect. |
