Objective: Through the establishment of a mouse model of Helicobacter pylori related gastritis(spleen deficiency damp heat syndrome),after the intervention of self-designed Qi Lian Wendan Decoction,the changes in the general condition of the mice,the eradication of Helicobacter pylori,the changes of the pathomorphology of the gastric mucosa,the expression of NF-kappa B p65 and I kappa B alpha in the gastric mucous membrane,and the treatment of the self prepared Qilian Wendan Decoction for the treatment of Helicobacter pylori were discussed.The possible mechanism of bacterial associated gastritis(spleen deficiency and dampness heat syndrome)is to provide experimental evidence for clinical treatment of Helicobacter pylori related gastritis.Methods: 60 SPF 6-8 weeks BalB/c mice were divided into 6 groups randomly,namely,model group(M),western medicine group(L),self-designed Qilian Wendan Decoction high dose group(WH),self-designed Qilian Wendan Decoction dose group(WM),self-designed Qilian Wendan Decoction low dose group(WL),blank control group(C).After 1 weeks of adaptive feeding,except for the blank control group,the other 5 groups were fed with self-made high fat and high sugar feed,and the animal model of the spleen deficiency and damp heat syndrome of Helicobacter pylori related gastritis was established by compound factor modeling method.After the model was successful,group WH,WM and WL were given high,medium and low doses of self-designed Qi Lian Wendan Decoction,and group II was fed with Li Zhu stomach triple suspension,M and C.Irrigate the stomach with equal volume of physiological saline.During the period of drug intervention,the mental activities,body weight,hair color,intake and intake of feces and feces of the experimental animals were monitored.After 14 days of drug intervention,the mice were killed and the eradication of Helicobacter pylori in the stomach of mice was observed.The pathological changes of gastric mucosa of mice were observed by HE staining.The expression of NF-kappa B p65 and I kappa B alpha in the gastric mucosa of micewere observed by immunohistochemistry.Results:(1)the mental state,hair color,activity,intake of water and stool in each dose group of Qilian Wendan Decoction group were better than those in the model group.(2)observation of Helicobacter pylori colonization in experimental mice showed that the M group was compared with the C group and found that the pyloric helicobacter pylori colonization score in the M group was higher than that in the C group(P < 0.01).Compared with the M group,the pylorus Helicobacter pylori colonization score in WH,WM,WL and L mice was lower than that of the M group(P < 0.05).(3)observation of the pathological morphology of gastric mucosa in the experimental mice showed that the M group and the C group were compared with the C group,and it was found that the grade of active inflammation and the reduction of the intrinsic gland in the M group were higher than that of the C group(P < 0.01),and the grade of active inflammation in WH,WM and L group was lower than that of the M group(P < 0.05).(4)compared with group C,the content of NF-p65 protein in the gastric tissue of M group was significantly increased,with significant statistical significance(P<0.01).Compared with group M,WH group and L group rats stomach tissue nf-kappa B p65 protein content decreased obviously,has significant statistical significance(P< 0.01),WM,WL group rats stomach tissue nf-kappa B p65 protein content is reduced,statistically significant difference(P< 0.05).(5)compared with group C,there was a significant decrease in the content of the protein in the gastric tissue of the M group,which showed significant statistical significance(P<0.01).Compared with group M,L,WH group rats stomach tissue I kappa alpha B protein content increased significantly,predominate has significant statistical significance(P < 0.01),WM,WL group rats stomach tissue I kappa alpha B predominate protein content decreased,with statistical significance(P< 0.05).Conclusion:(1)the self-designed Qi Lian Wendan Decoction can improve thegavage of Helicobacter pylori related gastritis(spleen deficiency and damp heat syndrome)in mice with mental malaise,burnout,hair sparsess,less intake of water and the symptoms of dry stool.(2)the self-designed Qi Lian Wendan Decoction can reduce the colonization of Helicobacter pylori in the stomach of mice with Helicobacter pylori related gastritis(spleen deficiency damp heat syndrome),and improve the pathological morphological damage of gastric mucosa in mice.(3)Qi Lian Wendan Decoction can affect the expression of NF-kappa B p65 and I kappa B alpha on gastric mucosa of mice with Helicobacter pylori related gastritis(spleen deficiency damp heat syndrome).The possible mechanism of the treatment of Helicobacter pylori related gastritis is to inhibit the activation of NF-kappa B p65 caused by Helicobacter pylori infection by up regulation of the expression of I kappa B alpha on the gastric mucosa,thus inhibiting the inflammation of the NF-kappa B p65 caused by Helicobacter pylori infection.The occurrence of symptoms and reactions can reduce inflammatory injury,so as to achieve the purpose of treatment of Helicobacter pylori related gastritis.
