Objective: 1.To explore the replicating method of sleep deprivation model after cerebral ischemiareperfusion injury.2.Sleep deprived rats after cerebral ischemia-reperfusion injury as a model.To explore the sedative and hypnotic effects of electroacupuncture(EA)on sleep disorders after ischemic stroke by selecting acupoint "Baihui"(GV 20)and "Shenting"(GV 24).3.To explore whether electroacupuncture on GV 20 and GV 24 can regulate the contents of monoamine neurotransmitters and interleukin-1 beta in hypothalamus,so as to improve sleep disorders after ischemic stroke.Methods: 1.12 healthy adult male SPF grade Wistar rats were randomly divided into the control group and the model group.The rat model of cerebral ischemia-reperfusion injury was reported by Zea Logna.After successfully replicating the model,the rats in the model group were injected with parachlorphenylalanine(PCPA)to prepare the sleep deprivation model after cerebral ischemia-reperfusion injury.After 7 days of observation,enzyme linked immunosorbent assay(ELISA)was used to detect the contents of monoamine neurotransmitters about 5-hydroxytryptamine(5-HT),5-hydroxyindole acetic acid(5-HIAA),Dobaamine(DA),norepinephrine(NE)and interleukin-1?(IL-1?)in hypothalamus.2.Experimental method of sedative and hypnotic effects of EA on sleep deprived rats after cerebral ischemia-reperfusion injury: 12 Male adult Wistar rats were taken to build a rat model of sleep deprivation after cerebral ischemia-reperfusion injury.The model rats were divided into model group and EA group by random number table method,with 8 in each group.Intraperitoneal injection of pentobarbital sodium in two groups of rats.The rats in the EA group were acupunctured GV 20 and GV 24 after the injection of 4min while model group rats were not treated.The two groups of sleep time were observed in order to compare the sedative hypnotic effect.3.Experimental method of therapeutic mechanism of Electroacupuncture on sleep deprived rats after cerebral ischemia-reperfusion injury: 18 Male adult Wistar rats were taken to build a rat model of cerebral ischemia-reperfusion injury.Then according to the random number table method,model rats were divided into the model group,the diazepam group and the EA group.The rats in the three groups were established model of sleep deprivation after cerebral ischemia reperfusion injury.The model groups did not do any treatment,while diazepam group intraperitoneally injected diazepam and EA group acupunctured GV 20 and GV 24.Each group was treated for 7 days.After every day's treatment,the behavior changes of rats were observed and the score was recorded.After 7 days of treatment,using ELISA to detect the contents of monoamine neurotransmitters about 5-HT,5-HIAA,DA,NE and IL-1? in hypothalamus.Result: 1.The symptoms of nerve function defect in the model group were obvious,and the circadian rhythm disappeared.At the meanwhile,compared with the MCAO rats,the expression of 5-HT in the model group was significantly lower than that in the control group(P < 0.01),the expression of 5-HIAA and IL-1 beta was decreased(P < 0.05).2.The sedative hypnotic effect of EA,compared with the model group,the duration of sleep time increased significantly in the EA group(P<0.01).3.The mechanism of EA therapy,compared with the model group,the expression of 5-HT,5-HIAA and IL-1? in the EA group increased(P < 0.05).The expression of DA and NE in the EA group and diazepam group decreased(P < 0.05).4.The score of nerve function defect in the EA group was lower than that of the other three groups(P < 0.05).Conclusion: 1.The model of sleep deprivation after cerebral ischemia reperfusion injury can be replicated by intraperitoneal injection of PCPA in rats with cerebral ischemia reperfusion injury model.2.Electroacupuncture of GV 20 and GV 24 have a sedative and hypnotic effect on sleep disorders after ischemic stroke.3.To electroacupuncture GV 20 and GV 24 to improve the sleep disorder after ischemic stroke by up regulating the expression of 5-HT,5-HIAA and IL-1 beta,down regulating DA and NE in the hypothalamus.4.Electroacupuncture of GV 20 and GV 24 can improve the state of neural function defect in rats.
