Study Of Hypnogenic Effect And The Mechanism Of Auriculo-acupuncture On Rats Of Post Stroke Insomnia

Objective:1.In sleep deprivation rats after cerebral ischemia reperfusion injury as post stroke insomnia model,with embedding auricular point by intradermal needle as auriculo-acupuncture treatment method,to observe the effects of auriculo-acupuncture on the sleep time of Pentobarbital sodium rats with sleep deprivation after cerebral ischemia reperfusion injury to demonstrate hypnogenic effect of auriculo-acupuncture on rats of post stroke insomnia.2.To observe the effects of auriculo-acupuncture on monoamine neuro-transmitter and IL-1β in the hypothalamus of rats with sleep deprivation after cerebral ischemia reperfusion injury to investigate the mechanism of auriculo-acupuncture in treating post stroke insomnia.Methods:1.Male Wistar rats of SPF were established middle cerebral artery occlusion reperfusion model(MCAO)by Zea Logna’s middle cerebral artery occlusion,then established sleep deprivation model by intraperitoneal injection of p-chlorophenylalanine(PCPA).12 model rats were divided randomly into model group and auriculo-acupuncture group,and injected intraperitoneally with Pentobarbital sodium,auriculo-acupuncture group rats were treated with auricular point Shenmen(TF4)by embedding intradermal needle.The sleep time of each group rats was recorded.2.24 MCAO model rats divided randomly into control group,model group,Diazepam group,and auriculo-acupuncture group.Except for the control group,the other 3 groups rats were established sleep deprivation model by intraperitoneal injection of p-chlorophenylalanine(PCPA).The diazepam group rats were treated with diazepam via peritoneal injection,and auriculo-acupuncture group rats were treated with auricular point Shenmen(TF4)by embedding intradermal needle.After 7 days treatment,the content of 5-HT,5-HIAA,DA,NE and IL-1β in the hypothalamus of rats were determined by enzyme-linked immunosorbent assay(ELISA)method.Results:1.Compared with model group,the sleep time of auriculo-acupuncture group rats was obviously prolonged(P<0.01).2.Compared with the control group,the content of 5-HT,5-HIAA and IL-1β decreased(P<0.05),the content of DA,NE increased(P<0.01)in the model group;Compared with the model group,the content of 5-HT and IL-1β increased(P<0.01),the content of DA,NE decreased(P<0.01)in Diazepam group and auriculo-acupuncture group.Conclusion:1.Auriculo-acupuncture has hypnogenic effect on sleep deprivation rats after cerebral ischemia reperfusion injury.2.The effect of auriculo-acupuncture for treating sleep deprivation rats after cerebral ischemia reperfusion injury can achieve through increasing content of 5-HT,5-HIAA and IL-1β,and decreasing content of NE and DA in the body hypothalamus.
