Purpose:To observe the deliverance level of autophagy-associated protein MAP1LC3 and proliferative antigen Ki-67 in hrHPV-positive cervical tissue of early squamous cellulate carcinoma,HSIL,and normal or diagnosed as non-specific inflammation,to preliminary analysis of the independent role and correlation of MAP1LC3 and Ki-67 in the occurrence and gradual progression of cervical squamous epithelial diseases,to further analysis of its association with clinical indicators and pathological traits in early squamous cellulate carcinoma(I-IIA phase),to ascertain whether there is a combined effect of autophagy and proliferation on the malignant transformation of benign cervical diseases and the occurrence and progression of early carcinoma.Methods:From January 2016 to January 2018,cervical hrHPV-positive early squamous cellulate carcinoma(I-IIA phase)30 cases,HSIL 30 cases and normal or diagnosed as non-specific inflammation 20 cases treated by cervical biopsy,circular electrocautery resection,tapered cold knife resection,radical surgery,and surgical resection of uterine fibroids or other benign lesions in the affiliated hospital of Yan'an University,were respectively separately called early squamous cellulate carcinoma group,HSIL group and control groups.The deliverances of MAP1LC3 and Ki-67 in three cervical paraffin specimens were detected by immunohistochemical SP method and BenchMark XT automated method,respectively;semi-quantitative scoring method was used to report the results of immunohistochemical staining;SPSS 22.0 statistical software was used to analyze the deliverance of the two factors in the three groups,the influence of the correlation,and the effect on the clinical parameters and pathological traits of the patients in the early squamous cellulatel carcinoma group.Results:1.Most of the high deliverance sites of MAP1LC3 are in cytoplasm and nucleus.The positive deliverance rate of MAP1LC3 gradually decreased from the control group,the HSIL to the early staged squamous cellulatel carcinoma:70.0%(14/20),63.33%(19/30),and 26.7%(8/30),the positive rate of the early squamous cellulate carcinoma group was smaller than that of the HSIL group and the control group(P value<0.017).2.Most of the high deliverance sites of Ki-67 are in the nucleus.The positive deliverance rate of Ki-67 increased gradually from the control group,HSIL to early squamous cellulatel carcinoma:15.0%(3/20),56.67%(17/30),and 90.0%(21/30).The positive proportion of early staged squamous cellulate carcinoma was greater than that in HSIL and juxtaposed group(P value<0.017),the positive deliverance proportion of HSIL was greater than that of control group(P value<0.017).3.The deliverance of MAP1LC3 and Ki-67 in early squamous cellulate cancerous tissues was not interrelated to the sufferer's FIGO stage,the cancer size,and the muscular invasion situation(P value>0.05).The level of tissue differentiation was related to Ki-67(?~2=6.02,P=0.04),but not to MAP1LC3.4.There was a weak negative correlation between the deliverance of MAP1LC3 and Ki-67 in cervical squamous epithelium.(r_s=-0.255,P value<0.05).Conclusion:1.With the progress of cervical pathological changes,the deliverance of MAP1LC3decreased gradually,while the deliverance of Ki-67 increased gradually,weakly deliverance of MAP1LC3 and strong deliverance of Ki-67 in early squamous cellulate cancerous group,suggesting that the decrease of autophagic activity may promote the proliferation of early squamous cellulate carcinoma.This finding provides a new starting point for further exploring the complex mechanisms of cellular autophagy in the malignant transformation of benign cervical diseases and the occurrence and progression of early cancer.2.The deliverance level of proliferating antigen Ki-67 is related to the tissue differentiation of early squamous cellulate carcinoma of the cervix.3.In cervical squamous epithelial pathological changes,there is a weak negative correlation between the deliverance of MAP1LC3 and Ki-67,and there may be interaction between the two to promote cellular cancerous transformation and early cancerous progression,this study may have accumulated preliminary clinical data for cervical cancer related studies of cellular autophagy and cellular proliferation.
