Objective:To study the expressions of HIF-1α、Ki-67and P27in normal cervical epithelia(NCE), cervical intraepithelial neoplasia(CIN) and squamous carcinoma of cervix(SCC) and to evaluate their relationship with clinical pathological features and to understand the role of them in squamous carcinoma of cervix and the correlation in them.Methods:The immunohistochemistry(S7P method) was used to detect the expression of HIF-1α、Ki-67and P27of20cases of normal control,70cases of CIN and40cases with SCC.Results:1、The positive rates of HIF-1α was5.00%in NCE,37.14%in the CIN,75.00%in SCC;Expression rate in SCC and CIN were obviously higher than that incontrol group, there were significant differences (P<0.01); The positive rates of HIF-1α was18.18%in the CINⅠ tissues,35.71%in the CINⅡ tissues,65.22%in the CINⅢ tissues, there were significant differences (P<0.01); The expression of HIF-1α was correlated with histological grade, clinical stages, metastatic lymph node of SCC(P<0.05); But was uncorrelated with age and the general form of the organization(P>0.05);2, The positive rates of Ki-67was0.00%in NCE,41.43%in the CIN,72.50%in SCC. Expression rate in SCC and CIN were obviously higher than that incontrol group, there were significant differences (P<0.01); The positive rates of Ki-67was24.24%in the CINI tissues,50.00%in the CINⅡ tissues,60.87%in the CINⅢ tissues, there were significant differences (P<0.05); The expression of Ki-67was correlated with histological grade, clinical stages, metastatic lymph node of SCC(P<0.05); But was uncorrelated with age and the general form of the organization(P>0.05);3、The positive rates of P27was100.00%in NCE,61.43%in the CIN,37.5%in SCC. Expression rate in SCC and CIN were obviously lower than that incontrol group, there were significant differences (P<0.01); The positive rates of P27was81.81%in the CINI tissues,50.00%in the CINⅡ tissues,39.13%in the CINIII tissues,there were significant differences (P<0.01); The exprssion of P27was correlated with clinical stages, metastatic lymph node of SCC (P<0.01); But was uncorrelated with histological grade, age and the general form of the organization(P>0.05);4、The expression of HIF-la and Ki-67in SCC were positively correlated (rs=0.679,P<0.01), There were a negtively conelation between the positive rates of Ki-67and the positive rates of P27(rs=-0.564,P<0.01)Conclusions:1、The significant increased expressing of HIF-la in the SCC is closely correlated with the in-vasion biological behaviour of the cervical cancer tissue. The expression of HIF-la will be the decision of the malignant and prognosis of SCC, and to provide the theory according to treatment of SCC,2、Ki-67has important action in the occurrence and development of SCC. Detection of the expression of Ki-67is provided with important clinical appliedvalue in understand the biological behavior and estimation prognosis of SCC.3、P27is related to the occurrence, differentiation degree and lymph node metastasis and prognosis of SCC. Detection of P27protein expression could provide referring criteria for its diagnosis, predictor of prognosis and clinical comprehensive therapy.
