Objective:To study the correlation of IQGAP1 and Rho C in normal cervix, high-grade squamous intraepithelial lesion and squamous carcinoma of the cervix tissue by detecting the expression level of IQGAP1 and Rho C. And to analysis the role and significance of IQGAP1 and Rho C in the occurrence and development of squamous carcinoma of the cervix. Methods: Immunohistochemistry(IHC) Envision was used to measure the expression of IQGAP1 and Rho C in 13 cases of normal cervical tissues, 16 cases of HSIL(including 6 cases of cervical intraepithelial neoplasia grade II and 10 cases of cervical intraepithelial neoplasia grade III), 45 cases of squamous carcinoma of cervix. Chi square test(X2)was used to analysis the expression of IQGAP1 and Rho C in different cervical tissues and their relationship with various clinical pathological parameters, and the correlation between the expression of IQGAP1 and Rho C in squamous carcinoma of cervix was tested by Spearman rank. The two-sided test was applied in all of these results, P<0.05 was statistically significant. Results: 1. IQGAP1 was mainly located in the cell membrane and cytoplasm. 2. The positive expression rates of IQGAP1 in normal cervix, HSIL and squamous carcinoma of cervix were 23.08%(3/13), 25.00%(4/16) and 82.22%(37/45), respectively, and statistical significance was existed among the three groups(X2=27.538, P<0.001). However, there was no statistical significance between normal cervix group and HSIL group(X2=0.981, P=1.000>0.05). And statistical significance was found between HSIL group and squamous carcinoma of the cervix group(X2=17.153, P<0.001).And statistical significance was found between normal cervix group and squamous carcinoma of the cervix group(X2=17.439, P<0.001). 3. In squamous carcinoma of the cervix group, the expression of IQGAP1 was significantly related with cancer’ s differentiation degree, parametrial infiltration, vascular infiltration, lymph node metastasis and depth of stromal invasion(P<0.05), however, no statistically significant difference was found in the patient’s age, FIGO stage and tumor size(P>0.05). 4. Rho C was mainly located in cytoplasm in different cervical tissues. 5. The positive expression rates of Rho C in normal cervix, CINand squamous carcinoma of the cervix were 23.08%(3/13), 56.25%(9/16) and 82.22%(37/45), respectively, and the expression level of Rho C was statistically significant among the three groups(X2=20.712, P<0.001). No statistical significance was found between normal cervix and HSIL group(X2=3.972, P=0.133>0.05). However, statistical significance was found between HSIL group and squamous carcinoma of the cervix group(X2=9.219, P=0.010).And statistical significance was found between normal cervix group and squamous carcinoma of the cervix group(X2=14.819, P<0.001). 6. In squamous carcinoma of the cervix group, the expression of Rho C in patients was significantly related with patients’ age, differentiation degree and lymph node metastasis(P<0.05), however, there was no statistical significance in the patient’s FIGO stage, parametrial infiltration, vascular infiltration, depth of stromal invasion and tumor size(P>0.05). 7. There was positive correlation between the expression of IQGAP1 and Rho C in squamous carcinoma of the cervix(rs =0.392, P =0.008). Conclusion: 1. The positive expressions of IQGAP1 and Rho C showed an increasing trend among normal cervical tissue,CIN and squamous carcinoma of the cervix. Both of them may be associated with the occurrence and development of squamous carcinoma of the cervix. 2. The positive expression of IQGAP1 in squamous carcinoma of the cervix with lymph node metastasis group, vascular invasion group, depth of myometrial invasion group and parametrial infiltration group were higher than the control group, but it was not associated with patients’ age, FIGO stage and tumor size. The expression of IQGAP1 showed a gradually upward trend in highly, moderately and poorly differentiated squamous carcinoma of the cervix, and the level of IQGAP1 protein expression may be related with poor prognosis of squamous carcinoma of the cervix. 3. The positive expressions of Rho C in squamous carcinoma of the cervix with high age group and group with lymph node metastasis were higher than the control group; but it was not associated with depth of myometrial invasion,vascular invasion, parametrial infiltrationand, tumor size of patients. The expression of Rho C showed a gradually upward trend in highly, moderately and poorly differentiated squamous carcinoma of the cervix, and the level of Rho C protein expression may be related with poor prognosis of squamous carcinoma of the cervix. 4. There was positive correlation between the expression of IQGAP1 and Rho C in squamous carcinoma of the cervix tissues.
