Background and objective: The pathogenesis of nasal polyps is unclear.Although there is no reletively unified understanding of its pathogenesis.In rencent yeas,the pathogenesis of nasal polyps has been studied.The role of genetics and genetic changes in the pathogenesis of nasal polyps has begun to receive more attention.The understanding of nasal polyps goes further.In many cases,the formation of nasal polyps and tumor cells has certain similarities in cell growth regulation and gene expression activation.In recent years,studies have found that the gene of FLG has a certain relationship with many diseases.The FLG gene has multiple mutation sites,and the mutation point has obvious ethnic and demographic differences.The gene mutation of the FLG causes the corresponding protein expression to decline to cause a variety of allergic diseases.At the same time,several studies have pointed out that FLG mutation are associated with fish scales,atopic dermatitis,allergic rhinitis,and asthma.Allergc rhinitis,nasal polyps and bronchical asthma are three common respiratory diseases.There is a certain association between these three diseases,And it often manifest itself in the presence or transformation of each Other.In this study,the correlation between mutation sites and nasal polyps was investigated in the test of R501 X and 3321 delA of FLG.Methods: Genomic DNA was extract from 48 NP patients which has been confirmed as nasal polyp,and 30 normal human controls.Two FLG gene mutation points(R501X3321delA)were detected by PCR.Polypropylene gel electrophoresis is tested,and then seguencing.Results:There were two cases with FLG gene c.3321 delA,4.17%(2/48),all heterozygous mutations,and the control group did not have this mutation.At the same time,the new mutation site of FLG was found in c.1711C>A,.The base sequence is from CAC?AAC.There were thirteen cases group,25.00%(12/48).Two of which were homozygous mutation,4.17%(2/48).Only one of homozygous mutation was found in the control group,3.33%(1/30).Conclusion : The FLG gene mutation is associated with the occurrence and thedevelopment of nasal polyps and may be one of the causes of nasal polyps.
