| Obgective By cyclophosphamide(CTX)create immunosuppressive chemotherapy mice model,and observe the effects of acupuncture and moxibustion on indexs of spleen and thymus of mice,test the content of Interleukin-6(IL-6),tumor necrosis factor-alpha(TNF-?)in the serum and the activity of acid phosphatase(ACP),lactate dehydrogenase(LDH)in the spleen tissue after acupuncture and moxibustion,from the perspective of immunosuppression,preliminary discussion acupuncture and moxibustion mechanism of repair of the CTX chemotherapy immune function in mice,the interpretation of acupuncture and moxibustion improve molecular biology mechanism of immunosuppressive mice caused by chemotherapy.Methods According to the different days of treatment,divided into group 3d and group 5d,each group of 40 rats,divided into batch buy SPF males KM mices(7 weeks old,22±2g),adaptive feeding for 3 days,according to the weight of the 2 groups were stratified randomly divided into blank group,model group,acupuncture group,moxibustion group,each group of 10,and each group mices were dyed and num-bered.In the blank group,9% Nacl was injected into the abdominal cavity for 3 day saccording to the dose of 0.02ml/g,and the other 3 groups were injected with CTX solution according to the dose of 100mg/kg/d,and the immunosuppressive model was replicated for 4 hours after the end of the drug injection.At the time of treatment,t-he daily timers were responsible for the fixation of the blank group and the model group,and no other treatment was performed.The acupuncture group and moxibustion group were treated with acupuncture and moxibustion at the four points of "Dazhui","GeShu","Shenshu" and "Zusanli" respectively.According to the section of different treatment for 3 days and 5 days respectively,consecutivly.The mices in each group were removed from the eyeballs and take blood on the next day of the third day and fifth day of treatment respectively,separated from the supernatant and stored in the refrigerator at-20? to determine the content of IL-6 and TNF-?.After caesarean section,the spleen and thymus were removed quickly,and the wet weight were recorded and then stored in the refrigerator at-80? to determine the activity of ACP and LDH.Finally,we collate all the experimental datas and use statistical software to a n-alyze and draw the conclusion.Results 1 Acupuncture intervention for 3 days,compared with the blank group,the other 3 groups of spleen index and thymus index were decreased,and the difference was statistically significant(P<0.05);compared with the model group,acupuncture group and moxibustion group of mice spleen index and thymus index were increased,there were significant differences(P<0.05);compared with the acupuncture group,the indexs of spleen and thymus are higher than the moxibustion group,no significant difference(P>0.05).After 5 days,compared with the blank group,the other 3 groups of spleen index and thymus index were decreased,there was significant difference(P<0.05);compared with the model group,acupuncture group and moxibustion group,the spleen index and thymus index increased,there was significant difference(P<0.05);compared with the acupuncture group,moxibustion group refers to spleen index and thymus index is higher,but the difference was not statistically significant(P>0.05).2 After 3 days of tratement by acupuncture and moxibustion,the expression of IL-6 in the other 3 groups were decreased,and there was a statistical difference(P<0.05).Compared with the model group,the expression of IL-6 in the acupuncture group and the moxibustion group increased,and the difference was statistically significant(P<0.05).Compared with the acupuncture group,the expression of IL-6 in the moxibustion group was higher,with no significant difference(P>0.05).After 5 days of intervention,compared with the blank group,the expression of IL-6 in the other 3 groups decreased and had statistical difference(P<0.05).Compared with the model group,the expression of IL-6 in the acupuncture group and the moxibustion group increased with statistical difference(P<0.05).Compared with the acupuncture group,the expression of IL-6 in the moxibustion group was higher,but there was no significant difference(P>0.05).3 After 3 days of tratement by acupuncture and moxibustion,compared with the blank group,the expression of TNF-? in the other 3 groups decreased,and there was a statistical difference(P<0.05).Compared with the model group,the expression of TNF-? in the acupuncture group and moxibustion group increased,and there was a statistical difference(P<0.05).Compared with the acupuncture group,the expression of TNF-? in moxibustion group was higher,and there was no statistical difference(P>0.05).After 5 days intervention,compared with the blank group,the content of serum TNF-? expression in model group and acupuncture group were lower,with statistical difference(P<0.05),the content of serum TNF-? expression in moxibustion group is reduced,but no statistical difference(P>0.05);Compared with the model group,the expression of TNF-? in the acupuncture group and moxibustion group increased,with a statistical difference(P<0.05).Compared with the acupuncture therapy group,the expression of TNF-? in moxibustion group was higher with no statistical difference(P>0.05).4 Acupuncture intervention for 3 days,compared with the blank group,the content of ACP decreased in the other 3 groups of spleen tissues,and there were significant differences(P<0.05);compared with the model group,acupuncture therapy group and moxibustion group.The content of ACP expression were increased,there were significant differences(P<0.05);compared with acupuncture group,the expression of low content of moxibustion group,no significant difference(P>0.05).After 5 days of intervention,compared with the control group,decreased expression of ACP in the other 3 groups in the spleen tissue,with statistical difference(P<0.05);compared with the model group,acupuncture therapy group and moxibustion group.The content of ACP expression were increased,there were significant differences(P<0.05);compared with the acupuncture group,the low expression of ACP in moxibustion group,with no significant difference(P>0.05).5 Acupuncture intervention for 3 days,compared with the control group,decreased expression of LDH in the other 3 groups of spleen tissues,there were significant differences(P<0.05);compared with the model group,acupuncture therapy group and moxibustion group.The content of LDH expression were increased,there were significant differences(P<0.05);compared with acupuncture group,moxibustion the content of LDH in the treatment group,the expression is low,and there is significant difference(P<0.05).After 5 days,compared with the control group,the expression of LDH in the other 3 groups were decreased,there was significant difference(P<0.05);compared with the model group,the content of LDH group and acupuncture moxibustion group in the expression were increased,there were significant differences(P<0.05);compared with acupuncture group,LDH content of moxibustion group in the group of low expression,and there is significant difference(P<0.05).Conclusion 1 After the intraperitoneal injection,CTX can decrease the index of spleen and thymus,causing severe damage to the spleen and thymus of mice.Acupun-cture and moxibustion can improve the index of spleen and thymus,promote the repair of spleen and thymus,and relieve immunosuppression.2 CTX can reduce the content of IL-6 and TNF-? in serum.Acupuncture and moxibustion can increase the content of IL-6 and TNF-?,promote the expression of IL-6 and TNF-?,activate the immune effect of B and T cells,improve the immunosuppression induced by chemotherapy and improve the immunity of the body.3 CTX can decrease the activities of ACP and LDH in the spleen.Acupuncture and moxibustion can increase the activities of ACP and LDH,accelerate the express i-on of ACP and LDH,promote the activation of phagocytes,repair and protect the da-maged spleen,antagonize the immunosuppression induced by chemotherapy and en ha-nce the immune function of the body.4 Acupuncture can repair damaged immune function and improve immune suppression after chemotherapy. |
