ObjectiveUsing Ultraviolet B(UVB) to irradiate the murine dorsal skins to set up models of skin immunosuppression. Obeserving three kinds of cyclooxygenase-2 inhibitors' protection effects on the density and morphology changes of epidermal Langerhans Cells (LCs) to explore the new theory of Nonsteroidal Anti-Inflammatory Drugs (NSAIDs) on prevention or treatment of skin tumors.Materials and MethodsFifty-six healthy BABL/c mice(half male and half female) were selected and grouped into seven groups by random. They are control group(Group C),topical flufenamate ointment given once per day group(Group Fl),topical flufenamate ointment given twice per day group(Group F2),topical flufenamate ointment vehicle given once per day group(Group Vl),topical flufenamate ointment vehicle given twice per day group(Group V2),celebrex oral administration group(Group Ce) and indomethacin oral administration group(Group In).All mice were irradiated on the dorsal skins by UVB to establish the models of skin immunosuppression. The differences were determined of the density and morphology changes of epidermal LCs of murine dorsal skin,which were evaluated by immunohistochemical techinique,in the groups treated either with or without different types of NSAIDs.Statistical ananlysis: Repeated Measures ANOVA. Results1 Density changes of epidermal LCs1.1 The density of epidermal LCs decreased after UVB irradiation in all groups, and the tendencies were similar, i.e., they reached the lowest point on the 3rd day and began to rise on the 5th day. But there were siginificant differences among all groups.1.2 Compared with Group C,the density of LCs in Group F1,Group F2 and Group Ce were higher on the 1st,3rd and 5th days.1.3 Compared with Group C,the density of LCs in Group In was only higher on the 3rd day.1.4 Respectively compared with Group V1 and Group V2,the density of LCs in Group F1 and Group F2 were higher on the 1st,3rd and 5th days.1.5 Compared with Group F1,the density of LCs in Group F2 was higher on the 1st and 5th days.2 Morphology changes of epidermal LCsThere were different changes of LCs' morphology after UVB irradiation in all groups. Group C,Group V1 and Group V2 had the most significant changes,and Group F1,Group F2 and Group Ce had the least changes.Group In was in the middle.ConclusionsNSAIDs can siginificantly inhibit UVB induced murine skin immunosuppression. There are more protection effects in the topical flufenamate ointment and celebrex oral administration groups,but less in the indomethacin oral administration group. Besides, topical flufenamate ointment given twice per day has more protection than that is given once per day.
