The peony seed was used as a raw material in this study,and an enzyme-assisted n-hexane extraction system was developed for extracting peony seeds oil.Based on the single factor experiment,response surface method was used to optimize the extraction process of peony seed oil.Meanw-hile the physical and chemical properties and composition of seed oil were determined.Then the effects of peony seed oil on the improvement of drosophlia cognitive impairment were studied.The main research contents were as follows:1.pH value,reaction time and reaction temperature of enzyme-assisted n-hexane extraction of peony seed oil were optimized.Based on the single factor experiment using the response surface method.The optimal extraction conditions were pH was 5.1,the enzymolysis time was 68 min,and the enzymolysis temperature was 50?.Repeated 5 experiments were carried under the optimal conditions of the verification experiment andthe average extraction yield of the peony seed oil was 38.2%.However,the peony seed oil extraction yield untreated by enzyme pretreatment was only 27.8%.The extraction yield of enzyme treated peony seed increased 37.4%compared with untreated peony seed.This verification experimental is in good agreement with the predicted values of the regression equation,and the extraction conditions are with high reliability.2.Acid value,iodine value,peroxide value,saponification value and residual organic solvent of peony seed oil were measured according to national standard(GB/T5530-2005?GB/T5532-2008?GB/T5538-2005?GB/T5534-2008?GB2716-2005).The detection results of physical and chemical indexes accorded with the national standard of cooking oil.The main fatty acid components of peony seed oil wereanalyzed using a GC/MS.The content of untreated fatty acids of the peony seed oil is at about 90%.The main components are linolenic acid and linoleic acid.The oil also includes palmitic acid,arachidic acid,linoleic acid,linolenic acid,stearic acid.Specially,linolenic acid content is very high,reached more than 59%.The main components of peony seeds oil treated and untreated by enzymatic pretreatment had almost no difference.3.Through the pavlov smell escape reflex experiment,the smell short-term memory ability and the ability of climb drosophila,the smell of short-term memory and climbing ability of deosophila were analyzed for the normal group,positive control group,model group,the peony seed oil group,respectively.Experimental results showed that the peony seed oil could significantly improve the ability of climbing drosophila with tau gene and improve the cognitive function of drosophila.4.The wild-type controls drosophila and AD drosophila model(drosophila with tau gene)were obtained by the Gal4/UAS system hybridization.Through the pathological experiments,the AD model of drosophila was identified.According to the experimental requirements,drosophilas were divided into 6 groups.There are normal group,model group,three doses of peony seeds oil groups(0.2%,1%,5%)and donepezil group(positive group).Contents of cAMP and AchE in the drosophila brain were determinded and the pathological changes were observed in the brain of drosophila fed with peony seeds oil for 30days.Experimental results showed that the peony seeds oil could reduce the drosophila with tau gene brain lesion cavity and neurofibrillary tangles.Peony seeds oil can decrease the cAMP level and inhibited the activity of AchE in the brain of drosophila.Then it can improve the cognitive impairment of drosophila.
