Peony seed oil had trophism and pharmacological activity,was considered as various nutraceuticals,also was considered as high quality cooking oil.Peony seed had pharmacological activity substances,such as paeoniflorin and paeonol,had significant research value in tumour,nervous system and cardiocerebral vascular system.In this research,we optimized the synthetical extraction of peony seed oil,paeoniflorin and paeonol,refined the extracted oil,and studied the inoxidizability of peony seed oil.To research the extraction sequence of peony seed oil,paeoniflorin and paeonol,there were 2 groups according to the extraction sequence.Researched the extraction ratio of the three substances comprehensively,the optimum extraction sequence was peony seed oil firstly,and then paeoniflorin and paeonol.The experiment adopted organic solvent extraction,on the basis of single factor experiment,used response surface methodology to design and optimize the extraction process of peony seed oil.The results showed that the optimum conditions of extracting peony seed oil were determined as extraction for 5 h at 57℃with a solid/liquid ratio of 1:11,and the particle size was 40,extraction solvent was petroleum ether.Used peony seed meal as materials after extracting peony seed oil,extracted paeoniflorin and paeonol by the method of ultrasonic-assisted,on the basis of single factor experiment,used response surface methodology to design and optimize the extraction process.The results showed that the optimum conditions of extracting were determined as extraction for 2.3 h at 74℃with a solid/liquid ratio of 1:16,ultrasound time was 60 min,concentration of ethanol was 70%,the paeoniflorin yield was measured as 2.804 mg/g,the paeonol yield was measured as 0.418mg/g.To refine the extracted oil,the steps of degumming,deacidifying and decoloring were set different gradients to perform orthogonal experiments and researched the results by Principal Component Analysis,explored the optimization of extraction process for peony seed oil.In the refinement of peony seed oil,the quality of refined oil will be optimum after adding 4%boiled water,8%alkali liquor and 2%atlapulgite respectively,the physicochemical indexes of peony refined oil was:phospholipid content 8.37 mg/100 g,peroxide number 2.48 meq/Kg,iodine number160.85 g/100 g,acid number 0.37 mg NaOH/g,the refinement rate of peony refined oil was 21.37%through charcoal column.Compared the oxidation resistance of peony refined oil,procyanidine,resveratrol and VC by determining the free radical scavenging activity of DPPH,superoxide anion,hydroxyl and Fe2+-TPTZ.The results showed that peony refined oil had better scavenging activity than procyanidine,resveratrol and VC on superoxide anion at low concentration;when the concentration was 6 mg/m L,peony refined oil had better scavenging activity on hydroxyl free radical;the DPPH free radical scavenging activity of peony refined oil was less than VC but greater than procyanidine and resveratrol;in the method of FRTP,peony refined oil had better scavenging activity than resveratrol and VC at low concentration significantly.
