Objective:Zhi Yan Xiao decoction ( ZYXD ) is composed of astragalus, membranous, Rhizoma Alismatis, Schizandra chinensis baill, Folium Nelumbinis, Salvia miltiorrhiza Bge, et al. ZYXD hold the effect of strengthening spleen and tonifying the kidney, dispelling dampness and removing phlegm, assisting vital Qi and promoting blood circulation. ZYXD is an effective concerted granule which composed by my teacher according to the long observation of experiment and clinic. ZYXD has the effect of adjusting metabolizability of fat, resisting steatoperoxid, improving hepatic function and anti-hepatic fibrosis that has shown good effects on the treatment of clinic steatohepatitis. We apply modern medical knowledge to design the experiment on the basis of the former accomplishment this time. We duplicated the animal steatohepatitis model with high fat diet, studied the therapy of different dose and different period of ZYXD on nonalcoholic steatohepatitis and in order to provide an effective formula to clinical treatment by way of observing the effects of ZYXD on the experimental steatohepatitis rat.Method: 93 Wistar male rats were randomly divided into 7 groups. The NASH models were induced with fat-rich diet which is compose of 15 percent lard oil, 8 percent egg powder and 2 percent cholesterol, The normal control group (n=10) were fed with normal diet. The NASH model group included 18 rats, and were fed with fat-rich diet.At the 4, 8, 12 and 18 week, one rat of the steatohepatitis model group sacrificed for pathological observation each time.At the end of the experimentation, the NASH model group were 14 rats.The rest groups were large and small dose ZYXD treatment groups, the Dong Bao Gan Tai ( DBGT ) treatment group, and Ti Du Hu Gan Fang ( TDHGF ) treatment group, each groups included 13 rats respectively.The ZYXD prevention group were fed with fat-rich diet, and began to be perfused stomach with the ZYXD after 4 weeks, they were treated with ZYXD ( 1g/ml) 10ml/kg everyday. The rats of the rest treatment groups were fed with fat-rich diet, and they were treated by perfusing stomach after the rats have been induced the model with fat-rich diet for 12 weeks. The large dose ( 2g/ml) and small dose ( Ig/ml) ZYXD treatment group were treated with ZYXD 10ml/kg respectively everyday, The Dong Bao Gan Tai (DBGT) treatment group were treated with DBGT 10ml/kg ( 0.042g/ml) everyday;and the TDHGF treatment group were treated with TDHGF ( Ig/ml) 10ml/kg everyday;The normal control group and the NASH model group were treated with sodium chloride 10ml/kg everyday.At the end of 24th week, all rats were hocused with 2% pentobarbitalum natricum and killed. The serum level of liver function ( ALT, AST, Alb, Glo ) and serum lipid (TCH, TG, HDL-C, LDL-C ) were detected respectively with auto biochemical analysis apparatus.The level of superoxide dismutase( SOD ), malondealde ( MDA ), glutathione ( GSH ) and hydroxyproline ( Hyp ) in liver tissue were detected respectively with spectrophotometer. The serum level of TNF- α and IFN-γ weredetected with ELISA. The liver histological changes was studied with pathological observation and the positive expression rate of TGF-β1 was studied with the immunity organizes chemistry method. All data were compared among the seven groups.Result:During the experimentation, the weight of the NASH model group increase quicklier than the normal control group.After the rats have been induced the model with fat-rich diet for 12 weeks, the weight of the NASH model group was heavier averagely by 21 percent than the normal control group. At the end of experimentation, the weight and the rate of liver weight of the NASH model group were lower than the normal control group ( P<0.01 ), ZYXD could obviously decrease the weight and the rate of liver weight by comparison with the NASH model group ( P<0.05 ), but the rest treatment groups had no significant distinction by comparison with NASH model group.
