Object:This topic is based on the previous experimental study Zhigui shuxin particles of phlegm turbidity to merge the effects of myocardial ischemia model rats and Zhigui shuxin granule on the anterior descending coronary artery ligation in rats caused by the influence of myocardial ischemia,by observing the Zhiguil shuxin particles for different types of blood stasis model rats blood agglutination and the influence of blood rheology,explore the related action mechanisms of Zhigui shuxin particles promoting blood circulation to remove blood stasis.Methods:1.The artery-venous bypass thrombosis method,through the comparison of the changes in the weight of the blood clots in the stream of the bypass,observe the effect of Zhigui shuxin granules on bypass thrombosis in animals.2.Adopting adrenaline and ice water to stimulate blood stasis factors for the rat model of blood stasis,observe the changes of whole blood viscosity and plasma viscosity of Zhigui Shuxin Granule on blood stasis model animals under different shear rates.3.Rat model of venous thrombosis was reproduced by ligation of inferior vena cava proximal end?Inferior vena cava and left renal vein bifurcation below?,to observe the effect of different doses of test drugs on the thrombosis?wet weight and dry weight of thrombus?and the variety of factors related to thrombosis?serum thromboxane and prostacyclin?at the same time?4 hours after ligation?.Results:1.Effect on arterial-venous bypass thrombosis results show,Zhigui shuxin particles high dose,middle dose and low dose three to dose group and positive control group in the body of the animal-venous bypass formation of thrombus wet weight and dry weight were less than model group?P<0.05?,the results showed that Zhigui Shuxin granule has a very significant inhibitory effect on the thrombosis induced by aortic-venous bypass in rats.2.Effects of epinephrine and ice water on blood stasis syndrome rat model results show,compared with model group,the plasma viscosity of Zhigui Shuxin Granule high dose group was lower than that of model group?P<0.05?,it is suggested that Zhigui Shuxin granule can reduce blood viscosity to a certain extent,improve blood circulation and improve the degree of myocardial injury;at the 3/S shear rate,the viscosity of whole blood of Zhigui Shuxin Granules in high and middle dose groups was lower than that in model group?P<0.05,P<0.001?;at 30/S shear rate,the whole blood viscosity of Zhigui Shuxin Granules in high,middle and low dose groups was lower than that in model group?P<0.05,P<0.01?;at 100/S shear rate and 180/S shear rate,the whole blood viscosity of Zhigui Shuxin Granules of high dose group was lower than that of model group?P<0.001?.at other shear rates,the whole blood viscosity of the animals in each group dipped to a certain degree,but there was no significant difference in statistics.3.the results of the influence of venous thrombosis showed,the thrombosis wet weight and dry weight of thunderstorm induced by intravenous injection of Zhigui Shuxin Granules were lower than those of the model group?P<0.05?.Compared with the model group,the TXB2 content of Qigui Shuxin granules in the high-dose group,medium-dose group,low-dose group and positive control group were significantly decreased,with the difference between medium-dose group of Qigui Shuxin granules and the model group was statistically significant?P<0.05?,while there was a significant difference between the low dose group of Qigui Shuxin granules and the model group?P<0.001?.Compared with the model group,the contents of 6-k-PGF?in the high-dose group,middle-dose group,low-dose group,and positive control group were significantly increased,but the difference was not statistically significant?P>0.05?.It shows that Zhiguii Shuxin Granule has a very significant inhibitory effect on thrombosis induced by ligation of the inferior vena cava in rats.Conclusion:The experimental results show,Zhigui Shuxin particles may inhibit arteriovenous bypass thrombosis and venous thrombosis,improve plasma and whole blood viscosity,up-regulates 6-k-PGF1?and down-regulates TXB2,thereby regulating the ratio of TXB2/6-k-PGF1?,inhibit platelet aggregation,improve myocardial injury and other local blood circulation play a protective role in myocardial injury.This provides a reliable theoretical basis for the clinical application of Zhigui Shuxin Granule in the treatment of coronary heart disease and angina pectoris,and its mechanism of action needs to be further studied.
