Purpose: Observation of Zhigui Shuxin granules on rats model of myocardial ischemic injury,to investigate the protective effect of Zhigui Shuxin granules on experimental myocardial ischemia and its mechanism.Method: Select standard lead II ECG normal Wistar 60 rats,weighing 220 + 20 g,were randomly divided into: sham operation group,model group,positive medicine control group,Zhi GUI Shuxin granules in high dose group,Zhi GUI Shuxin granules in dose group,Zhi GUI Shuxin granule low dose group,with 10 rats in each group.After 14 days,30 days after oral administration,the model of myocardial ischemia was established by ligation of the left anterior descending artery of the left coronary artery in fifteenth days.24 hours after ligation,ethylurethanm anesthesia,standard lead II ECG records;the animals were sacrificed,abdominal aortic blood extraction,centrifuged to obtain serum for standby;rat heart,formaldehyde fixed alternate.To observe the degree of myocardial ischemia were observed by ECG ST segment deviation MV number and the pathological section of myocardium,detection of myocardial enzymes?LDH,CK,AST?,inflammatory reaction mechanism?TNF,IL-2,IL-6?,antioxidant mechanisms?SOD,MDA?and anti apoptotic mechanism?Bcl-2,Bax?and other indicators Zhi GUI Shuxin granules effect on ischemic myocardial injury,and to explore the mechanism of myocardial injury and the protective.Result: 1,Histological detection of electrocardiogram and electrophysiological shows,Zhi GUI Shuxin granules can reduce rat ECG ST segment deviation MV number,reduce the degree of myocardial injury.2,histomorphological study showed that Zhi GUI Shuxin granules can reduce rat myocardial necrosis area and improve the degree of myocardial injury of rats.3,test data show that Zhi GUI Shuxin granules,give medicine group cTn-I content,the content of IMA were significantly lower than those in the model group and the difference has statistical significance,indicating that Zhi GUI Shuxin granules could reduce the myocardial ischemia in the rat serum cTnI,IMA,alleviate the degree of myocardial injury of rats.4,through the detection of rat enzyme of energy metabolism of Ca2+-ATP content,indicating that Zhi GUI Shuxin granules can significantly improve the CA2+-ATP content in the blood,improve myocardial damage and improve myocardial energy metabolism.5,anti-inflammatory reaction mechanism showed that Zhi GUI Shuxin granules prior to ligation of the left coronary artery descending branch of law caused by myocardial ischemia model rats serum tumor necrosis factor alpha and leukocyte mediated IL 2 have a reduced role.In a certain extent can improve myocardial ischemia.6,antioxidant mechanism research shows that Guangxi Shu heart particles can decrease the content of MDA,increase the activity of SOD,relieve myocardial injury.7 and the mechanism of anti myocardial cell apoptosis research shows that Zhi GUI Shuxin granules by lower levels of Bax protein in the rat myocardial cells and bcl-2 protein content and the ratio of Bcl-2 / Bax increased,to reduce the degree of myocardial injury of rats.Conclusion: Through the experimental study showed that Zhi GUI Shuxin granules on myocardial ischemia of rats caused by myocardial injury in certain effect in alleviating and its mechanism may be related to Zhi GUI Shuxin granules can reduce myocardial cell injury and enhance antioxidant capacity and anti myocardial cell apoptosis and anti-inflammatory response ability.The Zhi GUI Shuxin granule application in treating coronary heart disease and angina pectoris provides a reliable theoretical basis,its mechanism still need further study.
