Objective: to examine the expression levels of B7H3,CD31,and ?-SMA in surgical specimens of 268 patients with gastric cancer were,to detect the expression of B7H3 in gastric cancer tissues and interstitium,besides,investigate the relationship with vascular endothelial cells and fibroblasts in tumor stroma,and its relationship with prognosis and survival of gastric cancer patients;Methods:268 surgically resected specimens of gastric adenocarcinoma confirmed by pathology were collected and the expression levels of B7H3,CD31 and ?-SMA in gastric cancer tissues and interstitial tissues were detected by immunohistochemical staining.Statistical analysis of B7H3 in gastric cancer tissues was performed.The level of expression and the correlation with the expression of interstitial blood vessels and fibroblasts,combined with the patient's prognosis were analyzed.Results: B7-H3 was expressed in both tumor cells and mesenchymal cells.The prognosis of patients with tumor cell B7-H3 expression was poor(n=65,P=0.015).The prognosis of patients with B7-H3 expression in mesenchymal cells was different.Not significant(n=154,P=0.255).Immunofluorescence staining showed that B7-H3 was expressed on tumor cells,CD31-positive endothelial cells,and ?-SMA-positive fibroblasts.Among them,interstitial B7-H3 was almost expressed on ?-SMA-positive fibroblasts.The correlation analysis between CD31 and ?-SMA and pathological parameters of the patients showed that the mortality rate of patients with high expression of ?-SMA was significantly increased,and the expression of B7-H3 was significantly correlated with the expression of ?-SMA in both tumor cells and interstitial cells.Positive correlation.The prognosis of patients with B7-H3 and ?-SMA double positive was significantly worse than that of other types of patients(P=0.001,P=0.004).Conclusion: B7-H3 was expressed in both tumor cells and mesenchymal cells,and was expressed mainly in ?-SMA positive fibroblasts in interstitial cells.The combination of B7-H3 and ?-SMA can be used as an indicator of poor prognosis of gastric cancer patients.combination.
