Purpose:In this study,the pathological model of ulcerative colitis(UC)was replicated with dextran sodium sulfate(DSS).The symplectic,serum enzymology,and molecular biology methods were used to study the effect of Xinkaikujiang on ulcerative colitis.Bitter-down method for treating ulcerative colitis.Method:In the experiment,60 C57 mice were used as research subjects.The model of ulcerative colitis was replicated using dextran sodium sulfate(DSS).The mice were divided into normal group,model group,control group,Wumei pill group and Banxiaxiexintang group,Gancaoxiexintang group.The normal group was drinking water and eating normally and was not given any medications or irritation.The other groups were given 3% DSS solution free drinking 7d model,3 days before the model was given respectively to the treatment drug(10% saline,sulfasalazine tablets,Wumei Pills,Banxiaxiexintang Tang,Gancaoxiexintang)continuous Dosing for 10 days.Finally,fasting and water were prohibited and samples were collected.1 Measure the length of mice's colon,detect the serum SOD,IL-6,and MPO levels,and detect the iNOS,COX2 and NF-kB protein levels in the colon tissue by Western blot.2 HE staining was used to examine the pathological changes of the colon of mice.All data was analyzed using SPSS20.0software.Measured data were expressed as mean±standard deviation(x±s).The data were tested for normality.The t-test was used for comparison between the two groups.One-way analysis of variance was used to compare data between groups;p<0.05 was considered statistically significant.Learn differences.Result:1 Mouse body weight: Compared with the normal group,the weight of the mice in the model group was significantly reduced(p<0.05);compared with the model group,the difference in body weight between the control group and each group was statistically significant(p<0.05);Compared with the control group,there was no significant difference between the Chinese medicine treatment groups(p>0.05).2 DAI score: Compared with the normal group,the DAIscores of the other groups were significantly higher.Compared with the model group,the DAI scores of the control group and the Chinese medicine treatment group mice were lower than the model group;compared with the control group,the Wumei pill group,The DAI scores of mice in Banxia Xiexin Decoction group and Gancao Xiexin Decoction group were lower than those in the control group.3 MPO: Compared with the normal group,the serum MPO levels in the model group increased significantly(p<0.05).Compared with the model group,the MPO levels in the mice of the control group and each group were significantly decreased.The difference was statistically significant(*p<0.05).Compared with the control group,serum MPO levels in the Chinese medicine treatment groups were significantly lower,and the difference was statistically significant(*p<0.05).4 IL-6:Compared with the normal group,IL-6 levels in the mice of the model group increased significantly(#p<0.05).Compared with the model group,the mice in the control group and each group were treated with serum IL.The content of-6was significantly lower,and the difference was statistically significant(*p<0.05).Compared with the control group,there was a significant difference between the Banxia Xiexin Decoction group and the Gancao Xiexin Decoction group(*p<0.05).There was no difference in the Wumei Pills group(p>0.05).5SOD:Compared with the normal group,the serum SOD content of the model group mice was significantly reduced,the difference was statistically significant(#p<0.05);Compared with the model group,the serum SOD content of the control group and the traditional Chinese medicine treatment group mice increased significantly The difference was statistically significant(*p<0.05).Compared with the control group,the SOD content in the mice in each Chinese medicine treatment group increased significantly,and the difference was statistically significant(*p<0.05).6 iNOS:Compared with the normal group,the iNOS protein content in the colon tissue of the mice in the model group was significantly higher(P<0.05).Compared with the model group,the iNOS protein in the colon tissue of the mice in the control group and each Chinese medicine group was significantly increased.The content was significantly lower and the differencewas statistically significant(*p<0.05).Compared with the control group,the iNOS protein content in the colon of Wumei Pill and Banxia Xiexin Decoction group decreased significantly,and the difference was statistically significant(*p<0.05),there was no significant difference in the decrease of Gancao Xiexin Decoction and it was not statistically significant(p>0.05).7 COX2:Compared with the normal group,the COX2 protein content in the colon tissue of mice in the model group was significantly higher(p<0.05).Compared with the model group,the COX2 protein content of the mice in each treatment group was significantly decreased,and the difference was significant.There was statistical significance(p<0.05).Compared with the control group,the contents of Wumei Pill,Banxia Xiexin Decoction and Gancao Xiexin Decoction group were decreased,and the difference was statistically significant(p<0.05).8 NF-kB: Compared with the normal group,the NF-kB protein content in the colon tissue of the mice in the model group was significantly higher(p<0.05);compared with the model group,the control group and the traditional Chinese medicine treatment group mice The content of NF-kB protein in colon tissue was significantly decreased,and the difference was statistically significant(*p<0.05).Compared with the control group,the content of Gancao Xiexin Decoction group decreased,the difference was statistically significant(*p<0.05),Wumei Pills The group and Banxia Xiexin Decoction group content decreased,the difference was not statistically significant(p> 0.05).9 Colon length of mice: Compared with the normal group,the length of the colon in the model group mice was significantly shortened,the difference was statistically significant(# p <0.05);compared with the model group,the colon length of the mice in the control group and each Chinese medicine treatment group was significantly longer The length of the model group was statistically significant(*p<0.05).Compared with the control group,the difference between the Chinese medicine treatment groups was not statistically significant(p>0.05).10 pathological examination: macroscopic observation of mouse colon tissue: normal group mice colon normal appearance,no congestion,normal color,serosal surface,mucosal surface and muscle layerno obvious lesions.The mice in the model group had part of the colon where the feces appeared to be full of intestinal tract,marked congestion,blood stasis,clear and thinning and shortening of the intestinal mucosa and muscle layers.The lesions in the control group and the traditional Chinese medicine treatment groups were significantly reduced compared with the model group.Compared with the control group,the lesions and colon morphology of the traditional Chinese medicine treatment groups were all reduced.The colonic tissue of mice was observed under a light microscope: In the normal group,the mucous membranes of the colon were intact,no inflammation was found under the mucous membrane,and the muscle structure was intact.In the model group,the mouse's colon loses its normal structure,inflammation of the colonic mucosa downstream,cell infiltration,colonic gland disappears,and is replaced by infiltrating inflammatory cells.The colon and mucous membrane inflammation,inflammatory cell infiltration in the control group and the traditional Chinese medicine treatment groups were all reduced compared with the model group.Compared with the control group,the colon structure and inflammatory cell infiltration of mice in the traditional Chinese medicine treatment groups were alleviated.Conclusion:Xinkaikujiang can achieve anti-oxidative stress and improve intestinal mucosal barrier function to achieve the purpose of treating UC.From the overall biochemical indicators,morphology and protein expression of colon tissue,the efficacy of each Chinese medicine treatment group was better than that of western medicine control group.In addition,in terms of Xiexintang and Wumeiwan,Banxia Xiexintang and Gancao Xiexintang focus on repairing mucosal lesions,reducing the area of ulcers,improving intestinal mucosal barrier function,and protecting mucosal integrity..Among them,Banxia Xiexintang is suitable for acute exacerbation,while Gancao Xiexintang is more suitable for chronic recovery period;Wumeiwan is more focused on anti-oxidative stress by inhibiting the iNOS-COX-2/NF-kB signal transduction pathway.Inhibition of IL-6inflammatory factors,increased SOD antioxidant capacity,play an anti-inflammatory effect,to achieve the purpose of treatment of UC.
