The Effect Of HuaZhuoJieDu YiKuiFang On The Expression Of IL-10 And ?-denfensin 2 In Rats With Ulcerative Colitis

Objective:By making the model of rats with ulcerative colitis (UC) and curing them, observed the variation of general state, intestinal mucosa with pathological changes, and the content of interleukin-10 (IL-10) and (3-denfensin 2 in them, discussed the mechanism of action of HuaZhuoJieDuYiKuiFang in curing ulcerative colitis, and offered the experimental proof for clinic.Methods:60 cleanly and healthy male Wistar rats were splited into 6 groups randomly according to random number table method:the blank group (A group), model group (B group), Mesalazine group (C group), HuaZhu-oJieDuYiKuiFang high dosage group (D1 group), middle dosage group (D2 group), low dosage group (D3 group), every of which containend 10 rats. Feeding normally for blank group, the rest of rats in every group were made for UC models via the method of trinitrobenzesulfonic(TNBS)/ethanol. Begaining with the fourth day after being made for UC models successfully, the rats of every group were fed normally, among 6 groups, the groups with treatment were given medical gavage on the basic of normally feeding, rats of C group were given the suspension of Mesalazine gavage on the basic of normally feeding, and rats of D1, D2 and D3 group were given different dosages of HuaZhuoJieDuYiKuiFang gavage. By the gavage of 2 weeks continuously, observing and recording the changes of the general state (spirit, coat, faeces, weight, activity, diet and so on) of rats, carrying on the DAI score. After finishing the treatment, every rat was stopped eating for 24 hours, but was not stopped drinking, we executed them by beheading and took the blood from them, testing the content of IL-10 in blood of rats in every group by radioimmunoassay, and dissected them so as to taking the colonic tissue. Then we carried the conventional paraffin embedding on the colonic tissue sectioned and processed HE staining, so that we could observed the situation of vitium, erosion and anabrosis in colonic mucosa, and detected the content of ?-denfension 2.Results:1 General stateA group rats were in good spirit. Others were weak, reacted slowly, ate lowly, lost weight, and their coat were dark yellow, faeces were rare or mixed up with mucus and purulent blood. However, after the treatment of medicine, the general state in rats of every treatment group was improved, activity and diet were increased, D1 group was the most. There was no obvious difference between C group ang D2 group, but both improvements were less than D1 group, but more than D3 group.2 DAI score resultsThe scores of C, D1, D2 and D3 group were lower than B group, difference was statistically significant (P< 0.05); the scores of C, D2 and D3 group were higher than D1 group, difference was statistically significant (P< 0.05); the scores of C and D2 group were lower than D3 group, difference was statistically significant (P< 0.05); the score of C group was not obviously different from D2 group, there was no statistically significant (P>0.05).3 Intestinal mucosa pathological observationThe intestinal mucosa of A group rats was normal. B group rats mucosa ulcered most, and existed obvious erosions, congestions and edemas, and the gland structure was destroyed. D1 group rats intestinal mucosa was better, and contained a small number of shallow ulcerations and new glands. C and D2 group rats mucosas owed smoothly, they were infiltrated by inflammatory cells, and small ulcerations were visible. The ulcerations of D3 group rats mucosa were more, the gland structure owed clearly, and congestions combine edemas were existent.4 Index detection The content of IL-10The contents of IL-10 in B, C, D1,1D2 and D3 group were lower than A group, difference was statistically significant (P<0.05); the contents of IL-10 in C, Dl, D2 and D3 group were higher than B group, difference was statisti-cally significant (P<0.05); the contents of IL-10 in C, D2 and D3 group were lower than Dl group, difference was statistically significant (P<0.05); the contents of IL-10 in C and D2 group were higher than D3 group, difference was statistically significant (P<0.05); the contents of IL-10 in C and D2 group were not statistically significant (P> 0.05). The content of ?-defension 2The contents of ?-defension 2 in B, C, D1, D2 and D3 group were higher than A group, difference was statistically significant (P<0.05); the contents of P-defension 2 in C, D1, D2 and D3 group were lower than B group, difference was statistically significant (P<0.05); the contents of ?-defension 2 in C, D2 and D3 group were higher than D1 group, difference was statistically significant (P<0.05); the contents of ?-defension 2 in C and D2 group were lower than D3 group, difference was statistically significant (P<0.05);the contents of ?-defension 2 in C and D2 group was not statistically significant (P>0.05).Conclusion:HuaZhuoJieDuYiKuiFang could improve the general state and indestinal mucomembranous damagement of ulcerative colitis rats.The mechanism curing UC maybe related to the increasement of IL-10 and the decreasement of ?-defension 2, both of which made the inflammation be relieved and the mucosa be repaired.
