Objective: To study the expression of Runt-related transcription factor 2(Runx2)in serum of patients with fracture complicated with traumatic brain injury(TBI)and to explore its effect on fracture healing.Methods : 25 patients of fracture combined TBI and 21 patients of simple fracture,who meet the inclusion criteria are selected,blood samples were collected on the 3,7,14 and 21 days after injury;Sixteen healthy people were selected as normal control group and blood was taken once.The levels of Runx2 and ALP in sera of patients at different time points after injury were compared by ELISA and RT-PCR,Patients of fractures X-ray were followed up on 1 and 2 months after injury,to understand the fracture healing.Results: The serum ALP levels of simple fracture group and fracture combined with TBI group on 3,7,14 and 21 days were significantly higher than those in normal control group(P <0.05);Serum ALP levels on 7,14 and 21 days after TBI in fracture group were higher than those in simple fracture group,the difference was statistically significant(P <0.05).The mRNA relative expression of Runx2 on 7,14 and 21 days after injury in simple fracture group increased by(1.49±0.33)times,(2.69±0.71)times and(2.76±0.89)times respectively than those in normal control group,the difference was statistically significant(P <0.05);3,7,14 and 21 days after injury,the Runx2 mRNA relative expression of fracture combined with TBI group increased by(1.56±0.36)times,(2.47±0.52)times,(4.16±1.25)times and(3.98±1.07)times respectively than those in normal control group,the difference was statistically significant(P <0.01);The degree of Runx2 increase in fracture combined with TBI group was significantly higher than that in simple fracture group.1 and 2 months after injury,the X-ray film suggested that the volume of the callus in the fracture-combined TBI was larger than that in the simple fracture,and the fracture healing rate was fast.Conclusions: The levels of Runx2 and ALP in the serums of fractures complicated with TBI were significantly higher than those of simple fractures,the callus formation volume was large,the healing rate of fractures was accelerated,and even the formation of heterotopic ossification(HO)may be related to the high expression of Runx2 and ALP,speculates that Runx2 and ALP participate in and regulate the process of bone regeneration.
