Objective: To investigate whether serum levels of CGRP,NGF and IGF changes were the main cause of fracture healing,through the establishment of TBI with white rabbit tibia fracture model,then clear mechanism of three kinds of factors in promoting fracture healing for patients with clinical bone delayed union and nonunion provides the theory basis for effective prevention and control measures and methods.Methods: Choose 48 New Zealand white rabbits of either gender,were randomly divided into four groups(N = 12):Group A: blank control group,Group B: simple brain injury group,Group C: simple fracture group,group D:bone fracture and cerebral trauma.Testing different set of rabbit at different times(6 h,3 d,7 d,14 d,21 d and 28 d)cerebrospinal fluid and serum levels of CGRP ?NGF and IGF-1.Group C and group D animals were sacrificed at 1,4 and 8 weeks after modeling and observed at the right tibial fracture site by X—ray and to be measured the callus volume.Then it will be analyzed by the pathology image analysis that the number of new blood vessels,new bone trabecular width,trabecular bone area percentage and other related components.Results: X –ray showed that fracture healing was accelerated in group D compared with group C.Callus volume measurement and composition analysis found that the volume of callus,the bone trabecular width,the trabecular bone volume area ratio,the number of new blood vessels of the group D was significantly higher than the group C after 4 weeks and the difference was statistically significant(P<0.05).ELISA detection showed that TBI combined fracture model NGF,CGRP,IGF-1 in serum and cerebrospinal fluid was increased significantly than in the simple fracture group(P < 0.05),and three factors in CSF reached the peak ahead of time than serum Conclusion:1.Brain damage can promote healing of fracture 2.CGRP,NGF and IGF-1 should be the factors involved in promoting fracture healing 3.The mechanism may be that they were released into the bloodstream after brain synthesis and come into play.4.CGRP played a role of time ahead of NGF,IGF-1 in promoting fracture healing.
