Objective1.To determine the correlation beteween Single Nucleotide Polymorphism with circulating intact parathyroid hormone in maintenance hemodialysis patients.2.To explore the relationship between Single Nucleotide Polymorphism and bone mineral density and cardiovascular calcification.Methods1.The clinical data of 70 hemodialysis patients of Outpatient Department in our hospital from June 2014 to February 2015 were analyzed retrospectively.Record demographic data,previous medical history,use of drugs,dialysis record and routine lab test results(including complete blood count,crp,25(OH)D,iPTH,hcy,comprehensive metabolic panel and iron metabolism).Seven SNPs(rs1570669,rs1801725,rs4074995,rs4443100 rs612709,rs73186030,rs219779)genotypes were dectected using Time-of-flight mass spectrometry.Then we make a correlative analysis between SNP genotype and iPTH.2.The clinical data of 47 hemodialysis patients treated with PTX for SHPT were analyzed retrospectively.Collect demographic data,anamnesis,use of drugs,dialysis record,routine lab test results(including complete blood count,crp,25(OH)D,ipth,comprehensive metabolic panel and iron metabolism),bone mineral density at lumbar and left hip joint by DXA,assenment of abdominal aortic calcification detected by lateral abdominal X-ray and number of calicified cardiac valve calcification by Echocardiography.Twelve SNPs(rs10500783?rs1459015?rs2036417?rs219779?rs307247?rs307253?rs4074995?rs6127099?rs6254?rs6442307?rs7652849?rs9630182)genotypes were dectected using Time-of-flight mass spectrometry to genotype.Then we make a correlative analysis and multiple stepwise regression between SNP genotype and bone mineral density and cardiovascular calcification assessment.Results1.Of the 7 SNPS analyzed for the hemodialysis group,MAF of rs1801725,rs4443100,rs73186030,rs219779 is less than 5%,the other 3 SNPS(rs1570669?rs4074995?rs6127099)were not correlated with iPTH in hemodialysis patients.2.0f the 12 SNPS analyzed for the SHPT group,MAF of rs219779?rs6442307?rs7652849 is less than 5%.rs1459015 correlated with hip joint mineral bone density(r=0.340,P<0.05)and T score(r=0.351,P<0.05).rs1459015 was found to be the most important independent determinant of hip joint bone mineral density followed by gender and 25(OH)D in a stepwise multiple regression model.Conclusions1.We didn't find any SNP correlated with circulating iPTH.2.Rs1459015 polymorphism not only correlated with hip joint bone mineral density in SHPT hemodialysis patients but also is an important independent determinant of it.
