Fissistigma retusum belongs to the genus of Fissistigma in the Annonaceae family,mainly distributed in Hainan,Guangdong,Guangxi and Yunnan provinces.They growth in the altitude of 340 m to 2000 m area,many of them in the mountainous jungle.The plant resources in Hainan Island are abundant,and its chemical composition has not been reported.Under the guidance of the anti-rheumatoid arthritis activity,the roots of F.retusum.were extracted with 95% EtOH and then evaluated for its anti-rheumatoid arthritis activity.The results showed that the EtOAc extracts exhibited significant anti-rheumatoid arthritis activity.So we studied on chemical constituents and anti-rheumatoid arthritis activity of the stems of F.retusum for the first time,which will provide a theoretical basis for the development and utilization of F.Retusum.In addition,we summed up 55 natural aristolactam alkaloids that were isolated from plants reported in about 50 articles,and their pharmacological activities also were summarized.In this study,the fractions were purified by means of silica gel column chromatography,Sephadex LH-20,preparative thin layer chromatography and recrystallization etc.The structures were elucidated by spectroscopic methods and comparison with the literature.Thirty compounds(1-30)were isolated from the stems of the F.retusum,including including 15 alkaloids and 15 non-alkaloids..They were identified as: goniothalactam(1),aristolactam BIII(2),piperolactam A(3),aristolactam A III(4),aristolactam FII(5),velutinam(6),aristolactam A Ia(7),aristolactam B II(8),lLysicamine(9),micheline B(10),norcepharadione B(11),anonaine(12),N-cis-feruloyl tyramine(13),aurantiamide(14),N-trans-cinnamoyltyramine(15),syringaresinol(16),epiyangambin(17),Seartemin(18),kaempferol-3-O-?-L-arabinopyranoside(19),naringenin(20),aromadendrin(21),eriodictyol(22),(2R,3S)-5,7,3'-tri-methoxy-epicatechin(23),5,6,8-tri-methoxy-7-hydro-xylflavonone(24),1-Syringoyl-1,2-dihydroxyethane(25),cis,transabscisicacid(26),(S)-methoxy-(3,5-dimethoxy-4-hydroxyphenyl)et-hanediol(27),?-Hydroxypropiosyringone(28),(+)-glaberide I(29),and protocatechuic acid ethyl ester(30).Among them,compounds 9-11,15-22,24,25,27,29 and 30 were isolated from the genus Fissistigma for the first time.All compounds were isolated from F.retusum for the first time.All extracts and the isolated compounds were evaluated their anti-rheumatoid arthritis activities.The results showed that compounds 2 and 22 exhibited weak anti-rheumatoid arthritis activities.
