The Fissistigma oldhamii belongs to the genus of Fissistigma in the Annonaceae family. Because of widely use in folk medicine, and containing anti-inflammatory and anti-tumor components, the genus of Fissistigma plants are favored by scholars at home and abroad. In previous studies by scholars, these plants were found to cantain alkaloids, flavonoids, organic acids and other compounds, furthuremore, alkaloids and flavonoids exhibited significant anti-inflammatory and anti-cancer activity. But so far,it's lack of systematic in the studys of anti-rheumatoid arthritis in genus Fissistigma as a whole. However, the study on composition and activity of Fissistiga from Hainan was almost vacuity. In order to illustrating the medicinal material base of Fissistigma for anti-Rheumatoid arthritis, enriching the phytochemical and pharmacological data of Fissistigma, measuring the regional differences of of phytochemical and pharmacological in genus Fissistigma, and as a pathbreaker for study of Fissistigma capitatum and Fissistigma tungfangense which specificly grow in Hai Nan, We made an alkaloid isolation study and system isolation study on the chemical constituents of Fissistigma oldhamil growing Hainan.In this study, the stems of Fissistigma oldhamil were purified by silica gel column chromatography, Sephadex LH-20, preparative thin layer chromatography and recrystallization and semi preparative HPLC separation method, 30 compounds(1-30) were isolated and puried from the sterms of the Fissistigma oldhamil,including 22 alkaloids and eights non-alkaloids.The structures of these compounds were elucidated using comprehensive spectroscopic methods.These were identified as orientaline-N-oxide(1), reticuline-N-oxide(2), fisoldhamone A(3), lanuginosine(4)?liriodenine(5), norcepharadione B(6), anolobine(7), xylopine(8), Nmethylbuxifoline(9), norannuradhapurine(10), anonaine(11), nuciferine(12),isocorydine(13), asimilobine(14), laurotanine(15), 3-hydroxynornuciferine(16),isoboldine-?-N-oxide(17), aristolactam AIIIa(18), piperumbellactam A(19),goniopedaline(20), aristololactam BIII(21), salutaridine(22), oxyphyllenodiol A(23),dysodensiols D(24), dysodensiols E(25),1,10-seco-4?-hydroxy-muurol-5-ene-1,10-diketone(26), 15-hydroxy-?-cadinol(27),eudesma-11(13)-ene-4?,9?-diol(28), aromadendrane-4?,10?-diol(29) and scopoletin(30). Among them, Compound 1 and Compound 3 are novel compounds. Compounds2, 4, 9, 13, 15, 16, 17, 19, 20 and 22-30 were isolated from the genus Fissistigma for the first time.The biological assayes including antimicrobial and anti-RA activities were used to find active compounds. Compound 5 exhibited broad spectrum antibacterial activity, Compound 13 exhibited activity against E.coli with MIC values of 5 ?g/mL.Compounds 1 and 2 exhibited anti significant anti-RA activity.
