Objective:Xinjiang Uygur and Kazak ethnic groups unique populations in the region,The use of whole-genome genotyping technology,to build Xinjiang Uygur and Kazak population population genetic structure,by comparison between Uygur and Kazak population genetic structure of the population difference of type 2 diabetes susceptibility genes,genetic structure differences were found between the two groups the most obvious type 2 diabetes susceptibility genes,so to explain the Xinjiang Uygur and Kazak population differences in incidence of type 2 diabetes reference.Method:(1)collect Xinjiang Uygur and Kazak healthy non-anticoagulated blood samples a total of 286 parts of whole genomic DNA extraction experiment,get Uygur and Kazak genomic DNA samples were screened 50 Uighur DNA from the sample sample,50 Kazakh DNA samples for subsequent genotyping experiments.(2)the use of Affymetrix Genome-Wide Human SNP array 6.0 Xinjiang Uygur and Kazak groups for whole-genome genotyping experiments,and construct maps based on population genetic structure of the resulting data.(3)For use Fst FSTT between Uygur and Kazak population genetic structure were compared,looking for population genetic structure differentiation between two distinct populations with type 2 diabetes susceptibility genes.(4)the use of software Hapmix type 2 diabetes susceptibility genes ancestral population mixing ratio estimates.(5)Use iHS model for Uygur and Kazak groups,comparative analysis of natural selection signal.Result:(1)Comparison of Uygur and Kazak population genetic structure by Fst FST.Found between the two groups of population genetic structure of the genome of a lower degree of differentiation,differences in population structure is a smaller group of two individuals.Similar Genome exceed 63.4%of polymorphic allele frequencies.(2)The study concerns 27 type 2 diabetes susceptibility genes genetic structure of the whole,there are significant differences(P<0.05)in the genetic structure between Uygur and Kazak groups.(3)In Ethnic Minorities in KCNQ1,exist ADCY5,CDC123-MIR4480,TCF7L2 gene and population genetic structure NDFIP2-SPRY2 significant difference,where there is the most obvious difference between the population genetic structure of KCNQ1(FST=0.0864)gene.(4)Uighur and Kazakh crowd ancestors mixing ratio estimation results,POU5F1-TCF19 gene region in the mixing ratio of the apparent gap between the European ancestry populations.In this vulnerable region,Kazakh mixed European ancestry was 58%,higher than the proportion of mixed ancestry Uighur Europe48%,and much higher than the average Kazakh genome mixing ratio in Europe(>3SD).(5)natural selection model detected,Uygur ARL15,CDKAL1,CDC123-MIR4480,KCNQ1,NDFIP2-SPRY2,FTO gene by natural selection pressure and influence by iHS.The Kazakh in CDC123-MIR448,KCNQ1,NDFIP2-SPRY2,LYPLAL1,POU5F1-TCF19and HHEX-EXOC6 subject to natural selection pressure.Conclusion:(1)Xinjiang Uygur and Kazak population 27 with type 2 diabetes susceptibility genes overall level of population genetic structure significantly different(P<0.05).(2)KCNQ1,ADCY5,CDC123-MIR4480,TCF7L2 and NDFIP2-SPRY2 gene may be one of the most important genetic factors Uygur and Kazak population incidence of type 2 diabetes difference.(3)POU5F1-TCF19 natural selection pressure affects only the Kazak population,and is the most significant among populations vulnerable populations mixed ancestry regional differences,may be one of factors resulting in differences in the incidence among populations.
