Objective: In this study,endogenous gangliosides(GLs)in serum of patients with intracranial tumors was quantified based on liquid chromatography-tandem mass spectrometry(LC-MS/MS)approach.And the correlation between the radiotherapy effect and GLs content before and after radiotherapy was investigated in order to discover new tumor maker for evaluation of clinical curative effect in brain tumor.Methods: 43 patients with intracranial tumors who had definitely pathological or imaging diagnosis and underwent radiation therapy from March 2016 to November2016 in our hospital was selected,including 27 cases of cerebral metastatic carcinoma and 13 cases of glioma.All the patients were separated into two groups.Group A was patients with tumors,among which 27 cases were cerebral metastatic carcinoma and 7cases of glioma.Group B had 6 cases of glioma postoperative.All the patients had reached the therapeutic doses,i.e.,patients with brain metastatic carcinoma had 40 Gy and those with glioma had 54 to 60 Gy.Meanwhile,neither metastasis to new organs nor tumor progression of other parts of the body happened during radiotherapy.The morning fasting venous blood of all subjects were collected and processed to obtain serum samples before and after radiotherapy within one week.Serum samples were then preserved at-80 ?.After extraction and purification,the collected samples were subjected to LC-MS/MS analysis.Two kinds of endogenous GLs including GM3 and GD3 were quantified before and after radiotherapy.The results were compared to the changes of imaging diagnosis after 3 months.And the correlation between curative effect of radiotherapy and GLs content in serum was systematically evaluated.Results:1.The result of serum GLs content in group AGroup PR had 7 cases of cerebral metastatic carcinoma patients that GD3 and GM3 content decreased after radiotherapy.Statistical analysis showed that the changes of GD3 and GM3 content before and after radiotherapy were statistically significant(P< 0.05).9 cases were in group PD,7 cases of brain metastatic carcinoma patients had GD3 content increasing 1.34 to 3.97 times after radiotherapy,6 patients rise by 1.23 to 1.23 times,1 patient was slightly lower;after radiotherapy,GD3 and GM3 both rise by 1.5times in 2 cases of glioma patients.Statistical analysis the changes of GD3 and GM3 content before and after radiotherapy has statistically significant(P < 0.05).There was 18 cases in group SD,the changes of GD3 and GM3 content in 13 cases of brain metastatic carcinoma patients had no significant after radiotherapy;5 cases of glioma patients had GD3 and GM3 content in varying degrees of decline,except 1increased obviously.But the change of GD3 and GM3 content has no statistical significance before and after radiotherapy(P > 0.05).2.The result of serum GLs content in group AGroup B had 6 cases of glioma postoperative,GD3 content changes in 0.9 to 1.1times after radiotherapy,and GM3 content fluctuated at about 0.7 to 1.2 times.The change of GD3 and GM3 content has no statistical significance before and after radiotherapy(P > 0.05).Conclusions: The patients of glioma and brain metastatic carcinoma had GD3 and GM3 downward trend in effective radiotherapy group(PR group);GD3 and GM3 has a downward trend after radiotherapy In invalid radiotherapy group(PD group);The changes of GD3 and GM3 has no obvious regularity in patients who were stability(SD)and postoperative(group B)after radiotherapy.So the change of serum endogenous GD3 and GM3 contents is consistent with the effect of the patients after radiotherapy.
