With modern society developing fast,the pace of life has sped up,pressure of people has increased,the impact of emotion has been magnified and the incidence of depression has been increased,which are extremely harmful to human physically and mentally.Thus,it has important theory significance and practical application value to develop novel high-perfomance natural anti-depression product with low toxicity and small adverse effect.Hemerocallis fulva belongs to the hemerocallis of liliaceae,a tradition plant of our country which can be used as both food and medicine.It is reported that the extract of hemerocallis fulva has the effect of anti-depression,but the active constituent is unknown.This investigation studied the anti-depression effect of hemerocallis fulva and selected active site as hemerocallis fulva flavonoids by forced swim test(FST),tail suspension test(TST),antagonism reserpine experimentand other depression models.And acute experiments TST and FST were performed to investigate the anti-depression effect of hemerocallis fulva flavonoids as despair depression models and the optimal dosage range.The result showed hemerocallis fulva flavonoids could remarkably shorten the immobility time of TST and FST and had significant anti-depression effect to the mice despair depression models.The optimal dosage are 10mg/kg?40mg/kg.The investigation studied the mechanism of the anti-depression of hemerocallis fulva flavonoids by determining content of monoamine neurotransmitter,content of blood serum corticosterone,BDNF of hippocampus,MDA content of postnasus cortex and SOD vitality.The result indicated that hemerocallis fulva flavonoids increased the 5-HT and DA of rats prefrontal cortex and hippocampus,decreased the content of blood serum corticosterone,increased BDNF of hippocampus of rats,decreased content of cerebral cortex MDA and enhanced SOD vitality of rats cerebral cortex.Thereby,the anti-depression effect of hemerocallis fulva flavonoids may be produced by increasing 5-HT and DA,inhibiting excess accentuation of HPA,neurotrophy effect to hippocampus and protection of oxidative stress injuries.
