Total flavonoids(TF)as one of the effective components of Hemerocallis citrina baroni(H.citrina)has the function of boosting immunity,protecting liver,anti-inflammatory,anti-tumor and lowering blood pressure and falling hematic fat.At present,traditional extraction methods were used to separate TF from H.citrina,which has the disadvantages of a waste of time,low efficiency and pollution of environment.In recent years,people have being paying more attention to the ultrasonic extraction(UE)in separating natural plant ingredient.In liquid,ultrasound produce ultrasonic cavitation which results local high temperature and high pressure with shock wave,micro jet and free radicals coming into being,which can make the damage to plant cell wall and assist the material inside the cell in flowing rapidly and enhancing the extraction yield of natural production.Being compared to the traditional extraction methods,ultrasonic also has advantages of low extraction temperature,short extraction time,simple equipment operation and applicable range.Electric field extraction(EFE)has been popular in recent years and has applied to chemical separation.The principle of electric field extraction is that electric field affect cell membrane by electric field stimulation,electric fusion and electroporation,which produce irreversible damage on cell membrane so that target component flow rapidly and extration yield of natural production improve.In my paper,Ultrasound was synergized with electrostatic field to extract TF from H.critrina making use of breaking wall of ultrasound and breaking membrane of electrostatic field,which give full play to the advantages of both in the separation of natural products and improve extraction yield of TF from H.critrina.Under the conditions of UE and ultrasound-synergized electrostatic field(UEE)considering the electrostatic field voltage,ultrasonic power,extraction time,ethanol concentration,solid-liquid ratio and extraction temperature to the influence of TF yield.The results were as follows:with electrostatic field voltage raising,TF yield increased and then fell,when the voltage exceeded 7kV,yield was maximal.When ethanol concentration increased,yield increased significantly and then fell rapidly.When solid-liquid ratio increased to 25mL/g,the TF yield increased,then declined.With time flying,yield increased then declined with 30min.With the augmentation of the ultrasonic power,the TF yield first increased then reduced,the most suitable ultrasonic power was 700W.With temperature rising,TF yield increased the fell,but the influence of extraction temperature was not obvious.In single factor experiments,the TF yield extracted by UEE were higher than UE,it demonstrated the interaction between ultrasound and electrostatic field and strengthen of ultrasonic cavitation effect that enhances cavitation yield,which affecting TF yield.According to single factor experiments,significant independent variables of extraction were optimized by response surface methodology(RSM),and the optimal extraction conditions were as follows:electrostatic field voltage of 7kV,ultrasonic power of 500.17W,ethanol concentration of 70%,and extraction time of 20.02min,and the predictable yield was 1.583%.Under these conditions,the experimental result was 1.536%and relative error was 2.97%which is in the range of error.Being compared to UE and water extraction(WE),UEE not only increases the extraction yield of TF but also exhibits an excellent antioxidant activity.In order to further verify the interaction between ultrasound and electrostatic field,the ultrasonic cavitation yield was measured by iodine release method.The results showed that the release of iodine increased when ultrasonic field treat potassium iodide solution,and it is a proof of synergistic effect between ultrasonic and electrostatic field.When ultrasonic-synergized electrostatic field treated potassium iodide solution for 30min,the synergistic efficiency between the ultrasonic and electrostatic field maximize.Iodine release method has proved that the electrostatic field can promote ultrasonic cavitation yield,and ultrasonic cavitation is the main driving force for the separation of natural products.Electrostatic field,regarded as a random interference in the sound field,strengthen the cavitation effect,The TF yield can also be improved because of the enhancement of ultrasonic cavitation.
