As a famous-region crude drug of Gansu province,Radix Hedysari was contained in the root of Radix Astragali in ancient herbal works.It was used as Radix Astragali for a long time.Since the 1985 version of the Chinese Pharmacopoeia published,it began to be distinguished from Radix Astragali and listed as a single traditional Chinese medicine?TCM?.Modern researches have certificated that it contains a variety of types of chemical compositions.However the studies of standards for it's content determination are still blank.So this present study aims to establish a method for the determination of Radix Hedysari,and to supply the basis for the multi index quality control of Radix Hedysari.It has extensive pharmacological efficacies,particularly has advantages in improving immunity,hypoglycemic and anti-oxidation efficacy.This paper expects to find and select other potential pharmacological activities by using modern technology within the guiding principle of traditional Chinese medicine theory.The study of TCM pharmacokinetic is one of the most important measures which can modernize the research and accelerate the globalism of TCM.It also has great significances in elaborating the drug mechanism and optimizing the production process as well as developing a reasonable dosing regimen.This article mainly from the following three aspects to study on Radix Hedysari:?1?To establish a HPLC method which was in good applicability and sensitivity to determinate the 8 index components of 24 batches of Radix Hedysari,included of adenosine,?-aminobutyric acid and six flavonoids?calycosin-7-glucoside,ononin,calycosin,isoliquiritigenin,formononetin,medicarpin?.Then the contents of the 8 components were selected as the factors for clustering analysis to looking for the optimum source.The results showed that the best quality of Radix Hedysari was come from Guanting town of Tanchang County in Longnan,Gansu.?2?To establish a HPLC method which has good accuracy and sensitivity to investigate the formononetin and medicarpin in rat plasma and tissues for studies on it's pharmacokinetics of the extract with ethanol of Radix Hedysari.The 2 constituents in plasma showed good linearity as well as in tissues,r2>0.9900,and the lowest quantitative detection line?LOQ?were defined at29.25 and 59 ng/mL.The RSD%of both precision and stability were measured below 15%,and the average recovery was between 85%and 105%.The half-life t1/2/2 of formononetin and medicarpin were 11.348 h and 0.454 h,respectively.The pharmacokinetic parameters showed that both they were in accordance with one compartment pharmacokinetic model,which means the drug will distribute rapidly after absorbed.?3?The rats was eccoped the both ovaries and intragastric gavaged with different polar solvent extracts of radix Hedysari.The effect of anti-osteoporosis then was discussed by mensurating the bone mineral densitometry of the ovariectomized rats and the indexes of mechanics of materials of it's right femur.Also the related serological indexes were detected after experiment.The test outcomes revealed it could significantly increase the bone mineral density and bone mineral content?p<0.01?in ovariectomized rats ig administration of ethanol extract of Radix Hedysari when compared with model group.It can improve the elastic modulus and the maximum load capability of right femoral,with a statistical difference compared to model group.The serum indexes including Ca?p<0.01?,P and AKP,showed a tendency of increase,while the STRACP was reduced observably?p<0.01?.So we could draw a conclusion that the ethanol extract of Radix Hedysari has a better therapeutic effect on osteoporosis induced by ovariectomy rats.
