The traditional Chinese medicine Hongqi(Radix Hedysari),which is a medicinalmaterial of Gansu famous region drug,is the dry root of Astragalus membranaceus(Hedysarum polybotrys Hand.-Mazz.)of legume polysequence rock.Medicinal materials of Radix Hedysari are rich in many types of chemical components,aiming at the extraction,separation and detection of chemical components in Radix Hedysari medicinal materials,so far it is still a hot and difficult point in research.In this study,solvent induced extraction,which is different from traditional pretreatment methods,can not only achieve the purpose of on-line enrichment and purification,but also operate simply and quickly.Moreover,using UPC2 as the means of analysis and Radix Hedysari medicinal materials as the entry point,the rapid detection,origin traceability and pharmacokinetics of chemical components in Radix Hedysari medicinal materials were studied based on UPC2 separation technology different from traditional RP-LC retention behavior.On the basis of the above analysis and practice,the retention behavior of flavonoids in Radix Hedysari in UPC2separation technology was systematically studied,and compared with the traditional UPLC,the advantages of UPC2 in the separation and analysis of traditional Chinese medicine were discussed,which provided new ideas and new analytical methods for the rapid detection,origin traceability,quality control and pharmacokinetic research of various chemical components in Hongqi medicinal materials.This study mainly from the following four aspects of the chemical composition of Radix Hedysari in a systematic and in-depth study:First,qualitative analysis of unknown components in Radix Hedysari based on solvent induced extraction-UPC2technology;The UPC2separation technology,which is different from the traditional RP-LC retention mechanism and separation effect,was used study Radix Hedysari medicinal materials,and the solvent induced extraction method was used to extract,purify and enrich Radix Hedysari medicinal materials online.Finally,three unknown chemical constituents,coumarin,vanillic acid and ferulic acid,were determined by qualitative methods such as standard reference substance and retention time.Secondly,the rapid detection and comparative study of 11 chemical constituents in Radix Hedysari;This study took Radix Hedysari medicinal materials as the research object,using HSS C188 SB chromatographic column,0.1%(v/v)methanol as modifier,gradient elution,and established a UPC2-PDA method for the determination of 11 chemical components in Radix Hedysari medicinal materials.The effects of SPE,solvent extraction method and solvent induced extraction method on the extraction effect of 11 chemical components in Radix Hedysari medicinal materials were investigated,and the complementarity of the two separation modes in the separation process was confirmed by comparing the UPC2 and UPLC in the separation mode.Third,based on the previous chapter,this study used UPC2technology with fast,low solvent consumption and different retention mechanism with traditional RP-LC to establish a new fingerprint of Radix Hedysari medicinal materials to obtain different fingerprint information of Radix Hedysari medicinal materials.At the same time,using 11 kinds of chemical composition content difference,principal component analysis and heat map of hierarchical cluster analysis,the origin traceability of 25batches of Radix Hedysari medicinal materials from different origin was studied.The results showed that the quality of 25 batches of Radix Hedysari medicinal materials from different origin in this study was the best in Minxian and Dangchang,Gansu Province.Fourth,this study selected solvent-induced extraction as a pretreatment method for plasma analysis.Using UPC2 separation technology,a rapid and sensitive detection method was established for 4 components in rat plasma,such as3-hydroxy-9,10-dimethoxypterocarpane,formononetin and calycosin,by examining the retention behavior of UPC2 different from traditional RP-LC and using trans-cinnamic as internal standard.the study showed that the concentrations of the three target compounds were in accordance with the two-compartment model.According to the T1/2/2 values of 3-hydroxy-9,10-dimethoxypterocarpane,formononetin and calycosin,the fastest absorption and elimination of calycosin can be obtained.3-hydroxy-9,10-dimethoxypterocarpane was the slowest to absorb and remove.Fifth,this study used the UPC2 separation technique to investigate the retention behavior of seven different structure flavonoids in Radix Hedysari medicinal materials by examining the stationary phase,modifier,pH value,elution gradient,back pressure,temperature and injection volume.the retention mechanism of seven different structure flavonoids was also discussed.the results showed that the chromatographic column and modifier had the greatest influence on the retention behavior of seven different structure flavonoids;the other parameters were not the main factors affecting the retention behavior,and could be optimized in subsequent experiments.The results not only provide a new research idea for the separation and analysis of flavonoids in other medicinal materials;The separation and analysis of other types of chemical constituents in Chinese medicinal materials can be used as reference.
