As a genuine medicinal materials of Gansu province,Radix Hedysari has been widely used since ancient times.While the study on chemical constituents and pharmacological action of Radix Hedysari gradually improved,medicinal using value and developing potential of Radix Hedysari have been revealed.Since the 1985 version of the Chinese Pharmacopoeia published,it began to be distinguished from Radix Astragali and listed as a single traditional Chinese medicine?TCM?.The effects of Radix Hedysari on improving immunity,antioxidation,antitumor and other pharmacological activities have been reported.Presently,there are less research about theraputic effect of Radix Hedysari on osteoporosis.As a disease with high incidence and high risk coefficient,osteoporosis has seriously affected the quality of life of the patients,and has become a popular research field.So the partial least squares regression?PLSR?analysis and the gray correlation analysis were used to explore the correlation between peak areas of HPLC fingerprints and efficacy.At last,we observe the effects of active ingredient of Hedysari Radix on the function of osteoblasts.This thesis is mainly studied in the following three parts:The first part mainly studied the pharmacological activities of Hedysari Radix on osteoporosis.Osteoporosis mode was established through ovary resection of female rats,to select the best part after performing intragastric administration with different extracts of Hedysari Radix by measuring bone mineral density?BMD?,related bone mechanics and serological indexes of rats.The serum indexes including Ca,P and ALP,showed a tendency of increase,while the TRACP was reduced observably.So we could draw a conclusion that The ethanol extract of Hedysari Radix was the most effective part.The anti-osteoporosis effect of different source of Radix Hedysari ethanol extracts on ovariectomized rat was studied.It is found that the ethanol extract of 10 batch of Radix Hedysari can improve osteoporosis to varying degrees in which Radix Hedysari from Micang mountain in Wudu has a better therapeutic effect.The second part mainly studied the correlation of HPLC fingerprint of Radix Hedysari and anti-osteoporosis effect.The fingerprints of ethanol extract of 10 batches of Hedysari Radix from different habitats were established by HPLC.And the partial least squares regression?PLSR?analysis the gray correlation analysis were used to explore the correlation between peak areas of HPLC fingerprints and efficacy.Nine common peaks of the active fraction were selected.Adenosine,calycosin and formononetin in ethanol extract of Hedysari Radix have been identified separately among which adenosine and calycosin were found to be positively related to the pharmacodynamic data,and formononetin showed the negative correlation with the effectiveness.The results indicate that the effect is related to various components,among which adenosine and calycosin made the contribution to the anti-osteoporosis effect.The third part mainly studied the the components of Radix Hedysari on effect of osteoblasts.The researchers extracted osteoblasts from the skull bone of the rat.Adenosine and calycosin were added in culture medium after inducing osteoblast.Research suggests that calycosin can increase ALP activity of osteoblast and the optimum concentration was 1×10-5 mol/L.But adenosine has no effect on osteoblasts.Also,the result suggest that calycosin in optimum concentration had significantly increased the number of CFU-FALP compared with control group which means calycosin can also stimulate the differentiation of osteoblasts in vitro.
