Objective By researching the transcriptional regulation of Jiedu Quyu Ziyin Recipe on ER??ER? and the effect of the recipe on the PI3K/AKT signal pathway,to explore whether the mechanism of treating SLE with Jiedu Quyu Ziyin Recpe is related to regulating the transcription of ER and interfering the PI3K/AKT signal pathway,Methods Two groups of wistar mice are fed with Jiedu Quyu Ziyin Recipe decotion and normal saline respectively to get serum containing Jiedu Quyu Ziyin Recipe(drug serum)and the blank serum,the activity of drug serum was determined using HPLC united with ESI/MS;MCF-7 cells grown in 6-well plates in vitro were randomly divided into five groups:(1)blank serum group;(2)drug serum group;(3)MPP+drug serum group;(4)PHTPP+drug serum group.E2 was added into each group with the final concentration of 3 ng/ml,after 24 hours,the expression of ER??ER??PI3K and AKT mRNA in MCF-7 cells were analyzed by Realtime-PCRResults The drug serum contained ferulic acid and isoferulic acid,the quality of the drug serum has been well controlled;aftert intervention for 24 hours,(1)Both of the expression of ERa mRNA in drug serum group and MPP+drug serum group decrease compared with blank serum group(P<0.01);the expression of ERa mRNA in MPP+drug serum group decreases(P<0.01)while PHTPP+drug serum group increases(P<0.01)compared with drug serum group.(2)Both of the expression of ER? mRNA in drug serum group and MPP+drug serum group increase(P<0.05orP<0.01)compared with blank serum group,the expression of ER? mRNA in PHTPP+drug serum group decreases(P<0.01)compared with blank serum group;the expression of ER? mRNA in MPP+drug serum group increases(P<0.01)while PHTPP+drug serum group decreases(P<0.01)co mpared with drug serum group.(3)Both of the expression of PI3K mRNA in drug serum group and MPP+drug serum group decrease corypared with blank serum group(P<0.01),the expression of PI3K mRNA in PHTPP+drug serum group increases compared with blank serum group(P<0.01);both of the expression of PI3K mRNA in MPP+drug serum group and PHTPP+drug serum group increase compared with drug serum group(P<0.01).(4)Both of the expression of AKT mRNA in drug serum group and MPP+drug serum group decrease compared with blank serum group(P<0.01),the expression of AKT mRNA in PHTPP+drug serum group increases compared with blank serum group(P<0.05);the expression of AKT mRNA in MPP+drug serum group decreases(P<0.01)while PHTPP+drug serum group increases(P<0.01)compared with drug serum group.Conclusion Firstly,Jiedu Quyu Ziyin Recipe could reduce the expression of ER? mRNA and increase the expression of ER? mRNA in MCF-7 cells.Secondly,in transcriptional level,Jiedu Quyu Ziyin Recipe could inhibit the the PI3K/AKT signal pathway which is related to estrogen.Thirdly,the regulation of Jiedu Quyu Ziyin Recipe on ER??ER? was related to interfering the PI3K/AKT signal pathway.
