The Research On The Regulation Of Jiedu Qu Yu ZiYin Fang On ER Alpha/Beta And Apoptosis Of T Lymphocyte

Object To explore the adjust function of JieDu QuYu ZiYin Fang on T lymphocyte apoptosis and ER α/β, preliminary probe into the mechanism of treating SLE.Method HPLC was used to determine the concentration of effective components ferulic acid and isoferulic acid in the JieDu QuYu ZiYin Fang medicated serum prepared previously. CCK-8and flow cytometry detection were used to determine JieDu QuYu ZiYin Fang medicated serum on influence on the activity of T cell growth and T cell apoptosis, respectively. Caspase-3and ERα/β mRNA level were detected by realtime PCR while protein expression were measured by western-blot. ERα/β trancription activity was determined via Dual-luc assay.Results HPLC can be used to determine both concentrations of ferulic acid and isoferulic acid in JieDu QuYu ZiYin Fang medicated serum, which were10.18±0.36μg/mL、6.41±0.25μg/mL、5.88±0.16μg/mL in treated high, medium, low ferulic acid group, respectively. And42.23±0.96μg/mL、32.28±0.66μg/mL、27.56±0.46μg/mL in in treated high, medium, low isoferulic acid group. JieDu QuYu ZiYin Fang medicated serum can significantly promote T lymphocyte visable and non-viable apoptosis (P<0.05). Effectively improved the expression and activation of Caspase-3, reduced the expression of mRNA and protein of ER α/β and transcriptional activity which had a dose-response relationship.Conclusion JieDu QuYu ZiYin Fang inhibited T lymphocyte proliferation, promoted the apoptosis of T lymphocytes; reduced the expression of ERα/β and transcriptional activity to achieve therapeutic effects. JieDu QuYu ZiYin Fang inhibited the ER signal pathway by down-regulating the mRNA and protein expression and the transcription activity of ERa/p, so as to achieve therapeutic effects of SLE.
