[Objective]To observe the effect of Ziyin Huoxue Jiedu recipe on the damage of human umbilical vein endothelial cells(HUVEC)induced by advanced glycation end products zhjd and inhibitors(AGEs)in advanced glycation end product receptor(RAGE)and mammalian target of rapamycin(mTOR)protein expression and nuclear factor kappa B(NF-?B)effect nuclear transfer,explore the Ziyin Huoxue Jiedu recipe for the protective effect and mechanism of the injury of vascular endothelial cells from the positive rate of nuclear transfer status of NF-?B and the expression of RAGE and mTOR.[Methods]Cultured HUVEC cells with 100mg/ml AGEs induced by 24 h,were randomly divided into 5 groups,one group is the model group,in addition to the model cell group,which were randomly selected three groups were added RAGE inhibitor FPS-ZM1,NF-?B inhibitor PDTC and mTOR inhibitor rapamycin(RAP),the remaining50mg/ml zhjd added a group of concentrated liquid.After intervention 12 h,the morphological changes of the cells in each group were observed by light microscope.The expressions of RAGE and mTOR were detected by Western-blot,and the nuclear transfer status of NF-?B was detected by immunohistochemistry.[Results]1.Observation under optical microscope,cells without AGEs before spindle shaped monolayer growth and mosaic arrangement do not overlap each other,the cell membrane obviously,cell cell fusion of the bright mirror under the layer of cobblestone like arrangement;the abnormal proliferation of AGEs cells with 12 h cells,the cell number increased significantly,the majority of cell deformation the cell membrane shrinkage,fuzzy contour,change rough like particles in cells;inhibitor group and Chinese medicine group was shown to reverse the damage effect,see spindle normal cells more rounded shaped cells decreased,overlap cell decreased,cell proliferation decreased,cell membrane shrinkage deformation decreased obviously,reduce intracellular rough like particles.2.Compared with the model cell group,the RAGE expression level,mTOR expression level and NF-?B expression level of HUVEC cells in each group were significantly lower,and the differences were statistically significant(p<0.01).[Conclusion]In AGEs induced endothelial cell injury,Ziyin Huoxue Jiedu recipe can reduce the expression of RAGE,NF-?B and mTOR,thereby reducing the damage of AGEs to endothelial cells.This more significant protective effect may be related to the blockade of RAGE,NF-?B and mTOR three signaling pathways in Ziyin Huoxue Jiedu recipe.This may indicate that Ziyin Huoxue Jiedu Decoction can inhibit the three signal transduction pathways of RAGE,NF-?B and mTOR.
