Study On The Effect Of The TCM With Function Of Invigorating Qi And Tonifying Kidney On Foxp3mRNA In Treg Cells And STAT5mRNA In Decidua Tissue In Abortion-prone Mice

Objective To study the effect of the traditional Chinese medicine(TCM)with function of invigorating Qi and tonifying kidney on the expression of foxp3mRNA in CD4+CD25+regulatory T cells(Treg)at spleen and foxp3mRNA、NF-κ BmRNA、STAT5mRNA in decidua tissue in abortion-prone mice during pregnancy.Methods CBA female mice were mated to DBA/2 male mice as an abortion-prone model or to BALB/c male as an normal model group,which were randomy divided into 6 groups:The negative control group(fed with normal saline),the positive control group(treated with CsA),the Chinese medicine High-does group(treated-with 48g/kg),the Chinese medicine does group(treated-with 24g/kg),the Chinese medicine Low-does group(treated-with 12g/kg),the normal model group.on the 9th day of gestation,the mice were sacrificed in batches to take fresh sleepn for ectracting Treg cells.On the 9th and 14th day of gestation,the mice were killed in batches and their decidual tissues were stored into the refrigerator by minus 80 degrees.Their Treg on fresh splenic tissue were isolated and purified by magnetic separator,The purity of Treg before and after separation,were identified by FCM.The expresstion of Foxp3mRNA in Treg were used by RT-PCR.The decidual tissues was used to detect the level of foxp3mRNA、STAT5AmRNA、STAT5BmRNA、NF-κB mRNA by.RT-PCR.Results The effect of different interventions on foxp3 in Treg at sleepn:on the 9th day of gestation,the expressions of foxp3mRNA in Treg at spleen in High-does group of the therapeutic groups were increased obviously,comparing with the result in negative control group(P<0.05).From the numerical,there was obvious trend among these therapeutic groups but no difference(P>0.05).The effect of different interventions on foxp3mRNA、STAT5mRNA、NF-κBmRNA in decidual tissues:the expressions of foxp3mRNA、STAT5mRNA in decidual tissues in the Chinese medicine High-does group and the Chinese medicine Low-does group were increased obviously comparing with the result in negative control group(P<0.05)while the expression of NF-κBmRNA were decreased obviously(P<0.05).Conclusion The traditional Chinese medicine with the function of invigorating Qi and tonifying kidney could increase the expression of foxp3mRNA in Treg at spleen as well as the expression of foxp3mRNA、STAT5mRNA in decidual tissues in abortion-prone mice and therefore it could keep the state of immune tolerance which was necessary for pregnancy,at the same time,it could also decrease the level of NF-κBmRNA in decidual tissues and eventually leaded to the final success of pregnancy.
