Objective: The aim of this experiment was to observe the effects of the Traditional Chinese medicine with function of invigorating Qi and tonifying kidney on diabetes-related cognitive dysfunctions of rat and to study the mechanism. Methods: According to the theory that the original pathogenesis of diabetes is caused by the disorder of the kidney, the kidney produces marrow and comes through the brain, the syndrome differentiation of brain and blood, the multiplex rat's models with impairment of diabetes brain, hyperhomocysteinemia and incomplete cerebral ischemia were used as experimental object. After curing them with Naoshenkang, which based on the traditional Chinese medicine theories of invigorating Qi and tonifying kidney, we tested their capability of learning and memory, electroneurophysiology, pathophysiology, biochemistry and molecular biology index and so on. Results: The results of the study showed that the invigorating Qi and tonifying kidney traditional Chinese medicine—Naoshenkang possessed many effects as follow: lowering the level of blood glucose and homocysteine, improving the brain circulation, enhancing the sensitivity of insulin,improving the insulin resistance and upgrading superoxide dismiutase level. It could also decrease MDA content, regulate monoamine neurotransimitters content in the cerebral tissue, up-regulate nitric oxide synthase and acetylcholinesterase and neural growth factor and IGF-1 expression in rat hippocampus. In addition, Naoshenkang accommodated the expression of apoptosis protein Bax expression, up-regulated anti-apoptosis protein Bcl-2 expression and the ratio of Bcl-2/Bax, inhibited apoptosis of neurons in the hippocampus, and cured cerebral ischemic injury. It is obvious that Naoshenkang can ameliorate the injury of diabetes rat cerebral function caused by the high level of blood glucose and homocysteine. Conclusions: The traditional Chinese medicine with effect of invigorating Qi and tonifying kidney can significantly improve the rat's neural conduction function and the ability of learning and memory, recover the injured cerebral function. The mechanism may be carried out by the effects of the multi-levels, multi-steps and multi-targets.
