Effect Of Test Prescription For Invigorating Qi And Tonifying Shen On Expression Of CD4~+CD25~+ T Cells,Foxp3 In Spleen Of Abortion-prone Mice And The Outcomes Of Gestation

Objectiv To study the effect of test prescription for invigorating qi and tonifying shen on expression of CD4 CD25 T cells,Foxp3 factor of CD4+CD25+T cells in spleen of abortion-prone mice during pregnancy and outcomes of gestation.Methods CBA female mice were mated with DBA/2 male mice to establish abortion-prone models,which were randomly divided into 5 groups,the high dose of Chinese medicine group,the medium dose of Chinese medicine group,the low dose of Chinese medicine group,the negative control group(fed with normal saline),the positive control group(treated with CSA),the empbryo resorbing rate was counted on the 14th day of gestation,and proportion of CD4+CD25+T cells and Foxp3 factor of CD4 CD25 T cells in splenic were analyzed by flow cytometry(FCM)on the 9th and the 14th day of gestation.Results ?The effect of different interventions on expression of CD4+CD25 T cells in spleen of abortion-prone lice durIng pregnancy:on the 9th day of gestation,comparing with the negative control group and the positive control group,only the high dose of Chinese medicine group had significant differences(P<0.01).The high dose of Chinese medicine group had significant differences with the medium dose of Chinese medicine group and the low dose of Chinese medicine group(P<0.01),and there were no difference between other therapeutic groups(all P>0.05).on the 14th day of gestation,comparing with the negative control group,only the medium dose of Chinese medicine group had significant differences(P<0.05).comparing with the positive control group,only the low dose of Chinese medicine group had significant differences(P<0.05).The medium dose of Chinese medicine group had significant differences with the high dose of Chinese medicine group and the low dose of Chinese medicine group(P<0.05),and there were no difference between other therapeutic groups(all P>0.05).?The effect of different interventions on expression of Foxp3 factor of CD4+CD25+T cells in spleen of abortion-prone mice during pregnancy:on the 9th day of gestation,comparing with the negative control group,all therapeutic groups had significant differences(P<0.01,P<0.05,P<0.01).Comparing with the positive control group,only the high dose of Chinese medicine group had significant differences(P<0.01).The high dose of Chinese medicine group had significant differences with the medium dose of Chinese medicine group and the low dose of Chinese medicine group(P<0.05),and there were no difference between other therapeutic groups(all P>0.05).On the 14th day of gestation,comparing with the negative control group,only the medium dose of Chinese medicine group had significant differences(P<0.05).Comparing with the positive control group,all therapeutic groups had no difference(all P>0.05).There were no difference between all therapeutic groups(all,P>0.05).?The effect of different interventions on the outcomes of gestation:On the 14th day of gestation,comparing with the negative control group,only the medium dose of Chinese medicine group had significant differences(P<0.05).Comparing with the positive control group,all therapeutic groups had no difference(all P>0.05).The medium dose of Chinese medicine group had significant differences with the low dose of Chinese medicine group(P<0.05),and there were no difference between other therapeutic groups(all P>0.05).Conclusion Test prescription for invigorating qi and tonifying shen can make CD4+CD25+T cells and Foxp3 factor of CD4+CD25+T cells in spleen of abortion-prone mice with high expression of,therefore it can keep the state of immune tolerance which was necessary for pregnancy,and trending to reduce the empbryo resorbing rate,providing a certain theoretical basis for the application of Chinese drugs for invigorating qi and tonifying shen.
