Objective Preliminarily study the mechanism of governor vessel superficial insertion by thick-needle for treating ischemic facial paralysis in rats,through observing the recovery condition of facial muscle function,in order to provide the experimental proof for thick-needle treatment.Methods Using arterial embolization method to build the ischemic facial paralysis,60 Wistar rats were divided randomly into 4 groups:thick-needle group,western medicine group,normal control group and sham-operation group,all groups were induced into ischemic facial paralysis model except sham-operation group.According to neurological function deficit score,we observed the symptoms of neurological deficits and improvement during the treatment,the rats in thick-needle group were punctured ShenDao point everyday for 4 hours after the model was build 1 day later;the western medicine group were given prednisone by gavage,and vitamin B12 everyday by intraperitoneal injection;the sham-operation group and normal control group without any treatment.Then randomly chose 4 rats in every group on the 3rd,7th,14th post treatment,and measured the expression of HIF-1? in related vascular tissue with Immunohistochemistry(IHC),and the serum levels about NO with Enzyme-linked Immuno Sorbent Assay(ELISA)?ET with Radioimmunoassay(RIA).Results ?Ischemic facial paralysis rats but sham-operation group display obvious symptoms of neurological deficit and the behavioral score between three and five points.The neurological deficit of thick-needle group,western medicine group and normal control group gradually increase.The neurological deficit of medicine and electro-acupuncture is obviously higher than normal control group in the same phase(P<0.05),and thick-needle group in lower than western medicine on 7th day(P<0.05),but higher on 14th day(P<0.05).?By determinating the expression of HIF-1?,each model group increases inordinately compared with sham-operated group(P<0.05),and more obviously on 3rd day in thick-needle group and western medicine group(P<0.05),and to the top on 7th day then slip down;compared with normal control group there is no statistical difference(P<0.05),except on 14th day.?By determinating the level of NO,thick-needle group and western medicine group increase inordinately compared with the sham-operated group on 3rd day(P<0.05),then it reduces on 7th day and compared with result there is no statistical difference on 3rd day(P<0.05).The NO content in the blood of each group is almost at the same level on the 14th day(P>0.05).?By determinating the level of ET,each model group increases inordinately compared with sham-operated group on 3rd day(P<0.05),and it is obvious lower in thick-needle group and western medicine group compared with normal control group(P<0.05),as the treatment progressed,the ET content reduces in the blood in hick-needle group and western medicine group,and there is no statistical difference compared with 3rd day(P<0.05).The ET content in the blood of each group is almost at the same level on the 14th day(P>0.05).Conclusion?Governor vessel superficial insertion by thick-needle can improve of neurological function on ischemic facial paralysis in rats.?Governor vessel superficial insertion by thick-needle can improve the expression of HIF-1 a in ischemia tissue.?Governor vessel superficial insertion by thick-needle can improve the level of NO in ischemia tissue.?Governor vessel superficial insertion by thick-needle can restrain the level of ET in ischemia tissue.?Governor vessel superficial insertion by thick-needle is a effective method to treat facial paralysis,the mechanism maybe through improve expression of HIF-1 a in tissue and regulate the level of NO?ET in blood.
