Objectives:Study the correlation between six subtypes of abnormal phlegmatic temperament type of vitiligo and HLA DQB1*0303,HLA DQB1*0601 and HLA DQB1*0602 allele.Methods:334 cases of whole blood samples from uyghur people was collected,including 183 cases of vitiligo patients and 151 cases of healthy controls;After extracting of genome DNA,the quality was detedted by agarose gel electrophoresis.HLA DQB1*0303,HLA DQB1*0602,HLA DQB1*0601 allele was detected by Polymerase Chain Reaction sequence specific primers(Polymerase Chain Reaction PCR-SSP);According to the electrophoretogram of PCR amplification products to determine whether a candidate gene in the genome DNA being genetic variation,and compare its distribution in abnormal phlegmatic temperament type of vitiligo,healthy people and six subtypes of abnormal phlegmatic temperament type in vitiligo.Results:1)Compared with healthy people,there was statistical significance(P=0.000)in frequency distribution of HLA DQB1*0303,HLA DQB1*0601 and HLA DQB1*0602 three candidate allele in patients with abnormal phlegmatic temperament type of vitiligo,the distribution of HLA DQB1*0303,HLA DQB1*0601 candidate genes in abnormal phlegmatic temperament type of vitiligo patients was obviously higher than HLA DQB1*0602 candidate genes;2)abnormal phlegmatic temperament type six different subtypes of vitiligo and HLA DQB1*0303,HLA DQB1*0601 two candidate genes correlated(P=0.017;P=0.002),the difference was statistically significant,and with HLA DQB1*0602 candidate genes-no correlation(P=0.53);3)HLA DQB1*0303 genotype of HLA DQB1*0602-degree of relative risk ratio than the OR value from big to small in turn as follows:3.494(astringency)and 2.593(mild),1.098(solidification sample),2.222(sweet),3.500(salty),0.480(sour),except acidity phlegmatic temperament type the rest are all risk factors for vitiligo.In HLA DQB1*0303 genotype of HLA DQB1*0601 genotype-compare the odds ratio for:0.614(astringency)and 0.723(mild),0.756(solidification sample),0.593(sweet),1.556(salty),0.675(sour)in addition to salty type phlegm type,the rest of the five risk factors are not vitiligo susceptibility;In HLA DQB1*0602 genotype of HLA DQB1*0601 genotype-compare the odds ratio for:5.668(astringency)and 3.588(mild),1.453(solidification sample),3.750(sweet),2.250(salty),0.711(sour),except acidity phlegmatic temperament type the rest are risk factors for vitiligo.Conclusions:1)HLA DQB1 allele may be related to abnormal phlegmatic temperament of vitiligo in uighur people in xinjiang,especially HLA DQB1*0303,HLA DQB1*0601 allele are the risk factors for abnormal phlegmatic temperament type of vitiligo in Uighur people,the phlegmatic temperament type population who carry the HLA DQB1*0303,0601 candidate genes have significantly higher risk of suffering with vitiligo than population who does not carry such a gene;2)HLA DQB1*0303,HLA DQB1*0601,HLA DQB1*0601 candidate genes increased the probability of suffering vitiligo in Uighur people whose abnormal phlegmatic temperament type in Xinjiang,HLA-DQB1 alleles may be associated with the susceptibility of vitiligo,they are the susceptibility genes of vitiligo;3)genetic background differences may exist in the susceptibility in the six subtypes of abnormal phlegmatic temperament type in vitiligo with uighur people in Xinjiang.
