Objective: By using the method of gene knockout,knockouting CCR3 gene of mice,to observe the animal phenotype and nasal mucosa pathological changes of experimental group and control group mice and to survey the granule protein of EOS in bone marrow,peripheral blood and nasal lavage,in order to research the influence to the EOS by konckout CCR3 gene,looking for possible new approaches for the treatment of allergic rhinitis.Methods: Knockouting specificity the CCR3 gene by the method of gene knockout,inducing mouse model of allergic rhinitis with ovalbumin(OVA),by use of reverse transcriptase polymerase chain reaction(rt-pcr)method to detect the EOS granule protein mRNA level in mice bone marrow,peripheral blood,nasal lavage.Using Western blot to detect the quantity of EOS granule protein in mouse bone marrow,peripheral blood and nasal lavage fluidResults: After sensitization by allergen,the rhinitis symptoms and nasal mucosa pathological changes of knockout CCR3 sensitization mice group compared with normal sensitization mice group significantly reduce.2,After sensitization by allergen,the expression mRNA of EOS granule protein and the quantity of EOS granule protein in mice bone marrow,peripheral blood and nasal lavage fluid of knockout CCR3 sensitization mice group compared with normal sensitization mice group significantly reduced(P < 0.05).Conclusions: By using the method of gene knockout to knockout CCR3,it suggests that the differentiation and development,also include migration and tissue infiltration of EOS is restrained by inhibit the expression of CCR3.It maybe a new choice of the trement of AR...
