Objective:By applying gene knockout technology,constructing CCR3 gene knockout mice model,to investigate its effect on proliferation,the mRNA expression level of granule protein and apoptosis of mouse eosinophils,giving a theoretical basis for targeting gene-therapy against allergic rhinitis.Methods: Using gene knockout technology called homologous recombination,constructing CCR3 gene knockout mice model.Then culture eosinophils from bone medulla of CCR3 gene knockout mice and normal mice 14 days.The proliferation and apoptosis of eosinophils was estimated by cells calculating and flow cytometric.Using Q-RT PCR to measure the the mRNA expression level of granule protein of eosinophils.Results:1?The every-two-day cells calculating result indicate that the eosinophils proliferation of experimental group was slower compared with control group on the 10,12,14 day(P<0.01).2?Comparing with control group,within 10ng/ml IL-5 or without IL-5,the eosinophils apoptosis rate of experimental group was higher(P<0.0001).3?The mRNA expression level of eosinophils granule proteins(ECP?EPO?MBP)were lower in the experimental group than the control group.(P<0.05)?Conclusions: By knocking out the CCR3 gene,the proliferation and maturity of eosinophils were inhibited,however the apoptosis was enhanced.It could be thought as a innovative therapeutic method of allergic rhinitis.
