The Role Of Interfeirng RNA Gene Silencing Of CCR3in Eosinophil Of Allergic Rhinitis In Mice

Objective: Use of in vitro synthesis of RNA interference to silence geneexpression of CCR3in eosinophils, specific inhibition of CCR3expression ineosinophils, blocking the path of Eotaxin/CCR3to konw the changes of eosinophilsCCR3and granule protein in the bone marrow, peripheral blood,nasal lavage fluid invivo. in order to understanding the role and impact of eosinophils in this process, findnew treatments of allergic rhinitis.Methods: The use of nasal topical administration, CCR3siRNA shuttle plasmid(pLVX-ShRNA2-mCCR3-1+2+3+4) effective for and specific to the inhibition ofCCR3gene expression obtained from in vitro experiments was used in the murineallergic rhinitis, the application of reverse transcriptase-polymerase chainreaction(RT-PCR) were detected by mouse bone marrow, peripheral blood and nasallavage fluid eosinophils CCR3and granule protein mRNA expression level.Results:①In the bone marrow, peripheral blood, nasal lavage fluid, theexpression of CCR3mRNA was lower in the CCR3siRNA treatment group than thePBS treatment control and siRNA treatment control group(P<0.05);②In the bonemarrow, peripheral blood, nasal lavage fluid, the expression of MBP, ECP, EPOmRNA was lower in the CCR3siRNA treatment group than the PBS treatment controland siRNA treatment control group(P<0.05).Conclusions: This study suggests a therapeutic approach the genes CCR3,through intranasal mucosal administration of target gene siRNA,can inhibit indevelopment, migration and infiltration of eosinophils in the animal model withallergic rhinitis, thereby reducing eosinophil degranulation, reduce the inflammatoryresponse of allergic rhinitis．...
