Antrodia camphorate are mainly distributed in Taiwan mountain area of China,and grows specifically in Cinnamomum kanehirai.It contains rich nutrition,such as polysaccharides,triterpene,sterols and other physiological active ingredients,which has many virtues of protecting liver,anti-tumor,anti-oxidation,immune regulation,detoxification,anti-inflammatory and other functions.Recent years,It has been become a hot spots on research,But with the Cinnamomum kanehirai are classified as rare species,and the growth cycle of solid culture is long,So the submerged fermentation technology of Antrodia camphorate is increasingly attended by people.There were mounts of agilawood material in the fermentation broth of Antrodia camphorate,and the volatile substances have great value for the industry of pharmaceutical,food and spices.First,we mainly studied on the aroma components of fermentation liquor and optimized the fermentation conditions for increasing the production of ?-benzene ethanol;Secondly,we tested the biological activities of small molecule substances in fermentation liquid.The main content of the paper are as follows:1.Different extraction methods were compared to assess the effects on aroma compounds in the fermentation broth of Antrodia camphorate,And aroma compounds which were extracted using different solvents and SPME were analyzed by GC-MS.These results showed that the solvent of n-pentane-dichloromethane(2:1)was better in this research,there were more than fifty compounds by n-pentane-dichloromethane(2:1).And these solvents can be got easily,so we choose n-pentane-dichloromethane(2:1)to extract the aroma compounds,and analyze the aroma compounds in different carbon and nitrogen source fermentation medium.Results showed that the aroma compounds were consisted of esters,alcohols,aldehydes,heterocyclic and a little of acids compounds.2.The fermentation conditions were optimized for improving the production of ?-benzene ethanol.The optimal culture medium were:50g/L glucose,5g/L yeast powder,0.1%K2HPO4,0.05%MgSO4,0.01%VB1,and the optimum cultural time was seven days at 26? in 120mL/250 mL with the shake flasks at 150r/min.The best inoculation volume was 20%.Under these conditions,the content of the product(P-benzene ethanol)was increased to 1.236g/L3.The fermentation broth of Antrodia Camphorate was extracted with petroleum ether,chloroform,ethyl acetate and n-butanol to get different extracting crude products,respectively.We got petroleum ether phase,chloroform phase,ethyl acetate phase,n-butyl alcohol phase.Results showed the crude extracts of fermentation broth had the biological activity of anti-cancer effect,immunoregulation,antioxidation and anti-aging in vitro.4.Based on the biological activity of anti-cancer effect and the weight of these extractions,we choosed chloroform part and ethyl acetate phase to isolate and purificate using silica gel.Then we obtained compound ?,compound ?,fraction?,and fraction?.The chemical structures of the compound ?(5-hydroxymethyl-2-furancarboxaldehyde)were determined by ESI-MS,1H-NMR and13C NMR.
