Along with the increasing living standard of people,the incidence of obesity is also climbing year by year.Currently,obesity has become the fourth largest medical problem and has become a global epidemic.Therefore,to seek a safe and effective way against obesity has become a task for scientists to work day and night.There are reports of tea reduce body weight,especially oolong tea which has a unique effect of beauty and slimming.But scientific reports are few.In present study,functional components from oolong tea were isolated and investigated about their antiobesity effect in rat fed with high fat diet.Then at genomics level,we screening and analyzed related genes and its pathways in order to make sure of the functional components and their mechanism.Mainly the topic includes the following contents:We succeeded in separating functional components from oolong tea.Then their purity were determined,the results are as follows:in total water extraction,neutral sugars,uronic acid content,polyphenols are respectively calculated as 17.22±0.29%in glucose,26.38±0.12%in D-galacturonic acid meter,26.87± 0.15%in gae.In tea polysaccharide,neutral sugars,uronic acid content and protein content are respectively calculated as 26.66±0.04%in glucose,32.88 ± 1.54%in D-galacturonic acid meter and 0.77±0.02%.In tea polyphenols,the polyphenols content was 70.65 ±0.0032&in gae.Rat obesity prevention model is namely kicking out the obesity resistant rats two weeks after a high-fat diet.At the same time,we fed the rats with functional components and compared with positive weight-reducing medicine for 6weeks.According to the results we can see the body weight and fat index of all the groups are extremely lower than model group except for both the dosages of tea polysaccharides.The morphological observation of rat epididymal fat and liver cells presented that each dose drug could reduce the size of the fat cells and the degree of fatty liver.Detect cholesterl,triglycerides,malondialdehyde and total superoxide dismutase in serum and liver homogenate,as well as the high-density lipoprotein cholesterol,low density lipoprotein and leptin in serum.From the results we can conclude that both the low dosage of tea polysaccharide and tea polyphenols could decrease the concentration of MDA and increase T-SOD.The compound group could decrease the concentration of TC,TG and LDLC,synchronously,increase the concentration of HDLC.Tea polyphenols low dosage group,tea polysaccharide low dosage group and the compound group had a better effect and therefore use the three groups along with normal group and model group,we launched the production of microarray.Then we screened differential expressed genes associated with lipid metabolism.From the Kyoto Encyclopedia of Genes and Genomes website,we searched for the pathways of those genes which are fatty acid biosynthesis pathway,steroid hormone biosynthesis pathway,biosynthesis of unsaturated fatty acids pathway,fatty acid elongation pathway,glycerolipid metabolism pathway,glycerophospholipid metabolism pathway.
