ObjectiveThis paper proposes to discuss the possible mechanism of crocin against AS,through the detection of cholesterol content in RAW264.7 cells induced by OX-LDL,and the activity of NF-кB pathway related inflammatory factors(IL-1 β,TNF-α,MCP-1)and NF-кB.MethodsDivided the RAW264.7 cells into normal control group(NC group),OX-LDL group(model group,M group),OX-LDL + low concentration crocin group(LD group),OX-LDL + medium concentration crocin group(MD group),and OX-LDL+ high concentration crocin group(HD group).Induction group and different concentration of drug group were treated for 48 h with 50μg/ml OX-LDL to establish the foamy cell model.Different mediums containing drugs were given according to groups,while complete mediums were given in the control group,and collected the supernatant and cells after 12 h,24h and 48 h.Then detected the cholesterol content in cells by enzyme colorimetry,IL-1 β,TNF-α,MCP-1by ELISA and NF-кB activity by Western blot.ResultsCompared with NC group,the content of cholesterol in MD group and HD group decreased significantly(P < 0.05),their differences were statistically significant.The time after administration had little effect on the same drug group,and there was no statistical difference between the TC content of the same drug group measured at different time points(P > 0.05).The content of inflammatory factors in each drug group were lower than that in group M(P<0.05),which had statistically significant.And with the increase of concentration,the content of inflammatory factors decreased gradually.Compared with NC group,NF-кB protein activity in M group was significantly up-regulated;compared with M group,NF-кB protein activity in each drug group were lower than that in M group,and with the increase of drug concentration,NF-кB protein activity decreased in a dose-dependent manner(P < 0.05),the difference was statistically significant.ConclusionsCrocin can significantly reduce the cholesterol content in foamy cells and related inflammatory factors,and it may be achieved by inhibiting the NF-κB signaling pathway.
