Objective:The Biomacromolecule are extracted from Indigotica Fort,which contain polysaccharide(about 60%),protein,bioactive peptide and glucoprotein and have a lot of bioactivity.At present,people mainly do research on the anti-inflammation and immunity enhancing of the Biomacromolecule from Indigotica Fort.We have not heard about the reports in relation to anti-obesity,Lipid regulation and anti-oxidation effect of Biomacromolecule from Indigotica Fort.The experiment aims to observe the anti-obesity,Lipid regulation and anti-oxidation effect of Biomacromolecule from Indigotica Fort on the high fat diet and discuss the mechanism of the activity.Methods:Before the experiment,according to the weight and the lipid level of the SD rats, 72 male SD rats(110g-146g) were randomly divided into 6 groups,each group with 12 rats,the groups are normal control group,model group,low-dosage experiment group, medium-dosage experiment group,high-dosage experiment group and Orlistat control group.Each rat in every group was administered with liquid of 1ml·100g-1 per day.In experiment groups,rats were administered with Biomacromolecule from Indigotica Fort at the dosages of 20mg·kg-1,40mg·kg-1 and 80mg·kg-1,while in Orlistat control group with Orlistat at the dosage of 60mg·kg and the normal control group and model group with the same volume of distilled water.The whole experiment lasted for 40 days, during this period,the rats in normal control group were given normal diet,whilst in other groups with high fat diet,the body weight of rats were measured every ten days; the amount of food which rats ate were recorded every 24 hours;the total cholesterol (TC),triglyceride(TG),low density lipoprotein(LDL) and high density lipoprotein (HDL) in serum were tested at the beginning of the experiment,the 20th day by Tail Clip and the 40th day by intracardiac puncture;at the last day of the experiment,rats were administered,famished for 12 hours,then in anaesthesia with intake of ether and killed,then their fat tissues in the abdomenons,kidneys and genitals were removed and weight,then calculate the ration between the weight of fat tissues and body weight of the rats.the fat tissues in genitals were fixed and stained,then sliced up and the size of the lipocytes were observed by microscope;the activity of lipoprotein lipase (LPL),hepatic lipase(HL),and superoxide dismutase(SOD) and the content of malonaldehyde(MDA) from the serum and liver of the rats were also tested.Results:Biomacromolecule from Indigotica Fort can significantly lower the body weight, lighter the weight of fat tissues of the high fat diet rats and increase the feed efficiency of them.Biomacromolecule from Indigotica Fort can significantly make the lipocytes of the obese rats become small and decrease the level of total cholesterol(TC),triglyceride (TG) and low density lipoprotein(LDL) in the serum of the high fat diet rats. Biomacromolecule from Indigotica Fort can also significantly increase the activity of lipoprotein lipase(LPL),hepatic lipase(HL),superoxide dismutase(SOD) and lower the content of malonaldehyde(MDA) from the serum and liver of the high fat diet rats.Conclusions:From the results we can see that Biomacromolecule from Indigotica Fort can reduce weight and lower blood lipid.The effect of weight reduction of Biomacromolecule from Indigotica Fort do not curb appetite,in contrast,they can improve it.Biomacromolecule from Indigotica Fort may have the effect of weight reduction by means of enhancing the activity of LPL,HL and SOD,lipid regulating and lowering the content of free radicals and malonaldehyde.However,we still do not know what exactly is the reason of the effect of weight reduction of Biomacromolecule from Indigotica Fort,it needs to do more research on it to know more.
