[Purposes:]The research of apoptosis factor PTEN and apoptosis inhibiting factor Bcl-xl's expressions in recurrent abortion decidual tissue in mice,and the preliminary discussion on the functional mechanism of the traditional Chinese medicine“Zhuyunningl”on cells'abnormal apoptosis and the recurrent abortion,provided reliable theory basis for traditional Chinese medicine prevention and the treatment of recurrent miscarriage.[Methods]1.The establishment of repeated natural abortion in mice and traditional Chinese medicine's intervention:using CBA/J female mouse and BALB/C male mouse to establish the model of normal blank group,CBA/J female mouse and DBA/2 male mouse to establish the model of recurrent abortion group,and then according to the order of pregnancy,divided the pregnant mice into blank group,model group,western medicine group(aspirin),low dose group of Chinese medicine,middle dose group of Chinese medicine and high dose group of Chinese medicine.In each group,there are 15 mice.Weighing the mother mice in each group,giving medicine to the mice in each group,and intervening them by giving different doses of medicine,setting the“Jianshuan”day as the zero day of pregnancy,and beginning to fill their stomach from the first pregnancy day,gastric formula:the dose of per kilogram of mice' body weight equaled the average dose of human weight multiplying by(the reduction factor of mice' dose/conversion coefficients of humans' dose).When the mice were in the eighth days of pregnancy,they would be killed.Observed embryo abortion rate in each group,and got the deciduas and villi tissue from mice.2.By using semi-quantitative PCR and immunohistochemical methods,BCL-xl's expressive conditions were tested in each group's pregnant mice decidual tissue.[Result]1.The establishment of repeated natural abortion in mice and traditional Chinese medicine's intervention:compared with the blank group,the success rate of model group mice' pregnancy reduced significantly and the difference had the statistical meaning(P<0.05);Compared with the model group,the success rate of mice' pregnancy increased obviously in high dose group of Chinese medicine,middle dose group of Chinese medicine and western medicine and the difference had the statistical meaning(P<0.05);The success rate of pregnancy in low dose group of Chinese medicine increased slightly,but compared with the model group,the difference had no statistical meaning(P>0.05);Compared with the western medicine group,the success rate difference of middle dose group of Chinese medicine had no statistical meaning(P>0.05).2.immunohistochemical experimental results showed that:Comparing the model group and the blank group,the staining intensity of Bcl-xl in mice deciduas decreased obviously;Compared with the model group,the staining intensity of Bcl-x1 in mice deciduas in high and middle dose group of Chinese medicine and western medicine group enhanced obviously;Compared with the model group,the staining intensity was basically similar to the low dose group of Chinese medicine;Compared with the blank group,the staining intensity was similar to the middle dose group of Chinese medicine.Comparing the model group and the blank group,the staining intensity of PTEN weakened obviously in mice deciduas;Compared with the model group,the staining intensity of PTEN in mice' deciduas enhanced obviously in high dose group of Chinese medicine.Compared with the model group,the staining intensity was basically similar to the low dose group of Chinese medicine;Compared with the blank group,the staining intensity of the middle dose group of Chinese medicine weakened.3.The experimental results of semi-quantitative PCR showed that:1).Compared with the blank group,the content of mRN A in Bcl-x1 decreased obviously in the model group mice deciduas and the difference had statistical meaning(P<0.05).2).Compared with the model group,the content of mRN A in Bcl-x1 increased obviously in the middle dose group of Chinese medicine and the difference had statistical meaning(P<0.05).3).Compared with the blank group,the content of mRNA in PTEN genes decreased obviously in the model group mice deciduas and the difference had statistical meaning(P<0.05).4).Compared with the model group,the content of mRNA in PTEN increased obviously in the high dose group of Chinese medicine,and the difference had statistical meaning(P<0.05).[Conclusions]We could get the results by studying animals' experiments:1.Through the anatomy and HE staining observation,it showed that embryos model of abortion mice in the experiment was made successfully,and there were obvious differences in the embryo survival rate in six different experimental groups.2.It could prevent and improve the symptoms of abortion in mice in curing mice recurrent miscarriage by using“Zhuyunning I"in high and middle dose group of Chinese medicine and western medicine.The Chinese medicine“Zhuyunning I”could decrease the abortion rate of the recurrent miscarriage mice model and could protect the fetus obviously.3.PTEN in the normal group and high dose group of Chinese traditional medicine have high expression in deciduas.So,PTEN gene may have nothing to do with abortion.Dydrogesterone Tablets has no effect on PTEN,expression level is low4.It promoted the expressions of the apoptosis inhibiting factor Bcl-x1 and tumor suppressor gene PTEN after the uses of traditional chinese medicine(TCM)high dose in the pregnant mice.It adjusted conduction balances of some certain signaling pathways in the recurrent abortion mice deciduas,reduced the abortion rate of embryos,thus played an effective role in maintaining pregnancy in pregnant mice.Traditional
