| Objective:In animal experiment,set PTEN and BCL-XL on the PI3K/AKT signaling pathway as parameter to observe the curing effect of Zhuyunning-I in recurrent abortion.Immunohistochemistry SP and PCR were used to detect the expression of PTEN and BCL-XL cytokines after drug interference with the aim to primarily explore the correlation between the expression of PTEN and BCL-XL and recurrent abortion.To provide basic research for the study of the whole PI3K/AKT signaling pathway related to recurrent abortion.Methods:Animal experiment:DBA/2 and CBA/J female rats were mated to form the URSA model which was recognized at home and abroad,90 CBA/J female rats were randomly grouped into high,medium and low dose of Zhuyunning-I groups,western drug(dydrogesterone)group and model group with 15 rats within,mated with DBA/2 male rats of a half in number.BALB/c male rats mated with CBA/J female rats were to form the blank group,which contains 15 rats.Pregnant rats of each group were irrigated with different dose of medicine since the first day for 8 days,which were put to death in the 9th day.Immunohistochemistry SP were to detect the protein content of trophocyte PTEN and BCL-XL of pregnant rats.PCR were to detect the mRNA expression level of trophocyte PTEN and BCL-XL of pregnant rats.Results:Prescription Zhuyunning-I can reduce the expression level of protein content of trophocyte PTEN of RSA rats;Prescription Zhuyunning-I can increase the expression level of mRNA content of trophocyte BCL-XL of RSA rats,in order to keep fetus and prevent the happening of recurrent abortion.Conclusion:1.PTEN and BCL-XL associated with the happening of RSA.They may be one of the mechanism being the cause of recurrent recurrent abortion.2.Prescription Zhuyunning-I can increase the BCL-XL expression level in trophocyte of RSA rats,and make the excessive apoptosis of cells being restrained;Reduce the PTEN expression level in trophocyte of RSA rats,prevent the excessive apoptosis of cells,make cells grow normally,so as to achieve the ultimate goal of preventing the happening of recurrent abortion. |
