Objective:To study chemical components of Aconitum leucostomum Worosch.and toxicity of cardiomyocytes with raw materials and chemical components for safe and effective use in clinic.Methods:1.Chemical components of A.leucostomum were separated and purified using chromatographic techniques,and their structure were identified by various spectroscopic methods.2.Using cell culture techniques of myocardial cells were observed the effective of raw materials and chemical components.3.Using the method of intraperitoneal injection of mice to determine LD500 of two chemical constituents in A.leucostomum.Results:1.Fifteen compounds have been isolated from A.leucostomum,and the structure of twelve alkaloids were identified as nine C18-diterpenoid alkaloids:Lappaconitine?2?,N-deacetyllappaconitine?3?,Leuconine?9?,6-O-acetyl-acosepticine?10?,Ranaconitine?11?,N-acetylsepaconitine?13?,N-deacetylranaconitine?14?,Finaconitine?15?,8-deoxylappaconitine?12?;three C19-diterpenoid alkaloids,Delvestidine?1??Anthranoyllycoctonine?4??Aconitilearine?8?.one alkaloid and two non-alkaloids were still identified.2.The toxicity of cardiomyocytes with raw materials and chemical components:Delvestidine>Anthranoyllycoctonine>pH=4 parts>pH=8parts>aconitine>N-deacetyllappaconitine>Ranaconitine>Lappaconitine>A.leucostomum.water extracted solution;3.The LD500 of Lappaconitine and N-deacetyl-lappaconitine were 9.58 mg/kg and 20.89 mg/kg respectively.(The intraperitoneal injection LD500 of aconitine were 0.33 mg/kg).Conclusion:1.8-deoxylappaconitine is a new alkaloid.Seven compounds which are Delvestidine,Aconitilearine,6-O-acetylaco-septicine,Ranaconitine,8-deoxylappaconitine,N-deacetylranaconitine and Finaconitine are isolated from A.leucostomum for the first time.2.There are much toxicity chemical compounds in A.leucostomum,and a dependency with toxicity of dosage.3.Compared with aconitine,Lappaconitine,N-deacetyllappaconitine had lower toxicity.
