Objective: To analyze the pharmacognosy, the contents of total alkaloid in Aconitumleucostomum Worosch. from different districts and investigate the acute toxicity andanticancer activity vitro body of A. leucostomum Worosch. to provide scientific basis forsearching out its quality, toxicity and anticancer activity functions. Methods:Characteristic and microscopic identification methods were used to identify A.leucostomum Worosch. The total alkaloids in different samples were isolated by Soxhletextraction combined with different pH value solutions and extraction method. Ultravioletspectrometer was used to analyze the contents of total alkaloids in all samples. Activities,toxic behaviors and weigh changes of mice were observed and recorded by dosingdifferent concentrations of A. leucostomum Worosch, and the value of LD50was calculatedusing Karber method. Using different extracts of A. leucostomum Worosch. to detect theinhibition of variety cancer cells vitro body by MTT method. Results: The features of A.leucostomum Worosch. is its roots exhibited strips, and contain characteristic class triangleor square sclereid, pasted granule and fiber bundle. The contents of total alkaloid in theroots of A. leucostomum Worosch from Gongliu County is higher than the other twocountries. Within the death peak time of two hours, it revealed that the activities of micereduced, breathing rate increased, opithotonus and other toxic symptoms, and the deathrate inclined with dose. The value of LD50was3.21g/kg (2.74~3.75g/kg). Andcompared with control group, the changes of organ coefficient of heart, liver, spleen, lungsand kidneys were within0.04%,0.60%,0.25%,0.22%and0.19%, respectively.Extraction parts1,3and4have better effect to inhibit cancer cell-K562. Conclusion: By analyzing pharmacognosy, the contents of total alkaloids, acute toxicity and anticancereffect of A. leucostomum Worosch. to provide scientific basis for its quality assessmentand safety, reasonable clinic applications.
