ObjectiveIn this study,we used proteomic Western Blot to identify the differential expression proteins of chronic hepatitis B(CHB)in patients with chronic hepatitis B(CHB)by early intervention with iTRAQ.The patients were treated with western medicine treatment group,western medicine treatment group and healthy The expression of differential protein in the control group was changed.In order to obtain chronic hepatitis B liver depression and spleen deficiency based on the characteristics of drug intervention in the expression of protein,to explore the efficacy of drugs and syndromes of protein expression changes between the law.And to provide the experimental basis for the study of the pharmacological basis,the curative effect evaluation and the protein molecular markers of the drug-induced target of chronic hepatitis B.Methods12 patients with chronic hepatitis B liver depression and spleen deficiency were randomly divided into Chinese medicine and western medicine treatment group,western medicine treatment group,adding healthy people to control.(PSMA6,PF4 V,PSMA7,GRP78)were detected by Western Blot.The results were compared with those of the control group(P<0.05).The serum levels of PSMA6,PF4 V,PSMA7 and GRP78 were detected before and after treatment.The gray value of the target protein obtained from each lane was compared with the corresponding albumin gray scale value,and the relative gray value was obtained.Then,the gray value of the target protein was calculated by imageJ software.The relative gray values of the normal group in the antibody and the relative gray values of the patient group were imported into the GraphPad Prism 5 software,resulting in a mean +/-SD chart.The gender,age and laboratory indexes of each group were analyzed by IBM SPSS Statistics V20.0 software.Results1.Compared with the healthy control group,the expression of PSMA6,PF4 V,PSMA7 and GRP78 were up-regulated in the western medicine treatment group and the western medicine group before and after treatment,the difference was statistically significant(P<0.05).2.The expression of PSMA6 and GRP78 protein was significantly lower than that of the control group(P<0.05).The difference between the two groups was statistically significant(P<0.05).There was no significant difference in the expression of PF4 V and PSMA7.3.There were no significant differences in the expression of PSMA6,PF4 V,PSMA7 and GRP78 in the treatment of chronic hepatitis B patients with liver depression and spleen deficiency before treatment and after treatment.4.The levels of ALT,TBIL and HBV-DNA were significantly lower than those before treatment(P<0.05),and the difference of AST was significantly higher than that before treatment(P<0.05)(P<0.05).There was no significant difference in AST and TBIL between the two groups before and after treatment(P<0.05).The difference of ALT and HBV-DNA between the two groups was not statistically significant Compared with the western medicine group,the HBV-DNA in the treatment group was lower than that in the western medicine group(P<0.05).There was no significant difference between the treatment group and the Chinese medicine group in the treatment group.ConclusionsIn this study,we showed that the expression of PSMA6 in chronic hepatitis B patients with liver and spleen deficiency was significantly higher than that in healthy control group(P <0.05).The expression of PSMA6 protein in the treatment group was significantly decreased,speculated PSMA6 was a marker of the potential efficacy evaluation of chronic hepatitis B deficiency.This study is important for the study of TCM syndrome differentiation,syndrome evaluation and drug action target of chronic hepatitis B with liver depression and spleen deficiency.
